Abstract

The aim of this study is to the roles of nasal lavage fluid levels of MUC5AC, goblet cell hyperplasia and ultrastructure of nasal mucosa following ambient PM2.5 exposure in a rat model of allergic rhinitis(AR).Female Sprague-Dawley rats were assigned randomly into 3 groups: a negative control group(NC group),an ovalbumin(OVA)-induced AR model group(AR group), and AR model group(ARE group) inhaled to PM2.5 at 200 μg/m³, 3 h/d, for 30 days. Nasal symptoms, levels of MUC5AC in nasal lavage fluid(NLF), were measured in each individual rat.Goblet cell hyperplasia were examined histologically with PAS-stained. Nasal mucosa tissue ultrastructure were observed by scanning electron microscope.PM2.5 significantly increased the number of sneezes, nasal rubs and the amount of nasal secretion in rats with AR.Statistically significant differences in MUC5AC levels and goblet cell hyperplasia were observed between the AR model exposure to PM2.5 and the AR model group.The nasal mucosa of AR model exposure to PM2.5 was disordered,lodged,assembled and twisted.Our data indicate that an increase MUC5AC level in NLF and the development of nasal goblet cell hyperplasia may provide some clues for determining the pathogenic mechanisms of AR by exposure to PM2.5.

本研究旨在探讨在变应性鼻炎(AR)大鼠模型中,环境颗粒物PM2.5暴露后鼻灌洗液中黏蛋白5AC(MUC5AC)水平、杯状细胞增生及鼻黏膜超微结构的作用。将雌性斯普拉格-道利大鼠随机分为3组:阴性对照组(NC组)、卵清蛋白(OVA)诱导的AR模型组(AR组)以及吸入浓度为200μg/m³ PM2.5、每天3小时、持续30天的AR模型组(ARE组)。测量每只大鼠的鼻部症状、鼻灌洗液(NLF)中MUC5AC的水平。采用过碘酸希夫(PAS)染色法进行组织学检查杯状细胞增生情况。通过扫描电子显微镜观察鼻黏膜组织超微结构。PM2.5显著增加了AR大鼠的喷嚏次数、蹭鼻次数及鼻分泌物量。在暴露于PM2.5的AR模型组与AR模型组之间,观察到MUC5AC水平和杯状细胞增生存在统计学显著差异。暴露于PM2.5的AR模型的鼻黏膜出现紊乱、倒伏、聚集和扭曲。我们的数据表明,NLF中MUC5AC水平的升高和鼻杯状细胞增生的发展可能为确定PM2.5暴露引发AR的致病机制提供一些线索。
