A novel Gram-stain-negative, rod-shaped marine bacterium, designated strain F02, was isolated from a marine saltern in Weihai, PR China. The cells of strain F02 were approximately 0.8-1.0×3.0-4.0 µm and motile by means of a polar flagellum. Strain F02 grew optimally at 33-35 °C, pH 7.5 and in the presence of 3.0 % (w/v) NaCl. Strain F02 showed oxidase- and catalase-positive activities. Phylogenetic analysis of 16S rRNA gene sequences revealed that strain F02 belonged to the family Halieaceae and exhibited 16S rRNA gene sequence similarities of 96.6 and 96.4 % to the type strains of Chromatocurvus halotolerans and Parahaliea mediterranea, respectively. The major cellular fatty acids of strain F02 were C18 : 1ω7c, C16 : 1ω7c, C15 : 0 and C18 : 1ω9c. The major polar lipids of strain F02 were phosphatidylglycerol, phosphatidylethanolamine and one unidentified aminolipid. Strain F02 contained Q-8 as the sole respiratory quinone. The genomic DNA G+C content was 58.4 mol%. The genome sequences of strain F02 and Chromatocurvus halotolerans DSM 23344 had an OrthoANI value of 70.5 %, and the average amino acid identity value between the two genomes was 62.6 %. The sequence similarity value between the rpoB genes of strain F02 and Chromatocurvus halotolerans DSM 23344 was 79.5 %. On the basis of polyphasic analysis, strain F02 represents a novel species in a new genus, for which the name Kineobactrum sediminis gen. nov., sp. nov. is proposed. The type strain is F02 (=KCTC 52616=MCCC 1H00224).
一株新型革兰氏阴性、杆状海洋细菌,被命名为 F02 菌株,从中国山东威海的一个海洋盐场中分离得到。F02 菌株的细胞大约长 0.8-1.0μm,宽 3.0-4.0μm,通过极生鞭毛运动。F02 菌株在 33-35℃、pH7.5 和 3.0%(w/v)NaCl 的条件下生长最佳。F02 菌株氧化酶和过氧化氢酶均呈阳性。16S rRNA 基因序列分析表明,F02 菌株属于 Halieaceae 科,与 Chromatocurvus halotolerans 和 Parahaliea mediterranea 的模式菌株的 16S rRNA 基因序列相似性分别为 96.6%和 96.4%。F02 菌株的主要细胞脂肪酸为 C18:1ω7c、C16:1ω7c、C15:0 和 C18:1ω9c。F02 菌株的主要极性脂为磷脂酰甘油、磷脂酰乙醇胺和一种未鉴定的胺脂。F02 菌株含有 Q-8 作为唯一的呼吸醌。基因组 DNA G+C 含量为 58.4mol%。F02 菌株和 Chromatocurvus halotolerans DSM 23344 的全基因组序列的 OrthoANI 值为 70.5%,两个基因组之间的平均氨基酸同一性值为 62.6%。F02 菌株的 rpoB 基因与 Chromatocurvus halotolerans DSM 23344 的序列相似性为 79.5%。基于多相分析,F02 菌株代表了一个新属中的一个新种,因此提议创建一个新属,命名为 Kineobactrum sediminis gen. nov.,sp. nov.。其模式菌株为 F02 菌株(=KCTC 52616=MCCC 1H00224)。
