Abstract

A novel Gram-stain-negative, rod-shaped marine bacterium, designated as strain O448, was isolated from the coastal area of Weihai, China (122° 14' E, 36° 54' N). Cells of strain O448 were non-motile, aerobic, approximately 0.4-0.6 µm wide and 1.5-2.0 µm long. Growth occurred at 20-40 °C (optimally between 33-37 °C), pH 6.5-8.0 (optimally at 7.0) and in the presence of 1.0-5.0 % (w/v) NaCl (optimally between 2.0-3.0 %). Phylogenetic analysis based on 16S rRNA gene sequence indicated that strain O448 belongs to family Rhodobacteraceae and exhibits 16S rRNA gene sequence similarities of 94.3, 94.1 and 93.8 % to the type strains of Pontivivens insulae, Halovulum dunhuangense and Maritimibacter alkaliphilus, respectively. The major cellular fatty acid was C18 : 1ω7c and the major polar lipids were phosphatidylethanolamine, phosphatidylcholine and phosphatidylglycerol. Strain O448 contained Q-10 as the sole respiratory quinone. The genomic DNA G+C content was 65.6 mol%. It is evident from phenotypic data and phylogenetic inference that strain O448 represents a novel species in a new genus, for which the name Oceanibiumsediminis gen. nov., sp. nov. is proposed. The type strain is O448 (=KCTC 62076=MCCC 1H00233).

从中国威海沿海地区(东经122°14′,北纬36°54′)分离出一株新型革兰氏阴性杆状海洋细菌,命名为O448菌株。O448菌株细胞无运动性,好氧,宽约0.4 - 0.6μm,长1.5 - 2.0μm。在20 - 40°C(最适温度为33 - 37°C)、pH 6.5 - 8.0(最适pH为7.0)以及1.0 - 5.0%(w/v)NaCl(最适浓度为2.0 - 3.0%)条件下生长。基于16S rRNA基因序列的系统发育分析表明,O448菌株属于红杆菌科,与海岛海活菌(Pontivivens insulae)、敦煌嗜盐菌(Halovulum dunhuangense)和嗜碱海洋杆菌(Maritimibacter alkaliphilus)的模式菌株的16S rRNA基因序列相似性分别为94.3%、94.1%和93.8%。主要细胞脂肪酸为C18 : 1ω7c,主要极性脂为磷脂酰乙醇胺、磷脂酰胆碱和磷脂酰甘油。O448菌株含有Q - 10作为唯一呼吸醌。基因组DNA G + C含量为65.6mol%。从表型数据和系统发育推断可知,O448菌株代表一个新属中的新物种,为此提出新属名为海洋泥杆菌属(Oceanibiumsediminis),新种名为威海海洋泥杆菌(Oceanibiumsediminis weihaiense)。模式菌株为O448(=KCTC 62076 = MCCC 1H00233)。
