Hepatoprotective effects of silymarin against diclofenac-induced liver toxicity in male rats based on biochemical parameters and histological study.

Abstract

Diclofenac (DIC) is a phenyl acetic acid derivative which is well known for its analgesic and anti-inflammatory. In our study, the rats were divided into four groups. Group 1, control group; Group 2 received DIC-only; Groups 3 and 4 received DIC plus silymarin. The results showed that levels of CAT, SOD, GPx and GSH significantly reduced and levels of ALT, AST, ALP, total bilirubin, nitrite content, MDA, serum TNF-α and gene expression were significantly elevated in second group compared to control group. In other hand, treatment with silymarin resulted in a significant elevation in CAT, SOD, GPx, GSH and a significant reduction in MDA, ALT, AST, ALP, total bilirubin, nitrite content, serum TNF-α, and gene expression of in comparison with second group. Histopathological injuries were also improved by silymarin administration. The results confirm that silymarin has a protective effect on DIC-induced liver toxicity and oxidative stress in male rats.

双氯芬酸(DIC)是一种苯乙酸衍生物,以其镇痛和抗炎作用而闻名。在我们的研究中,将大鼠分为四组。第一组为对照组;第二组仅接受双氯芬酸;第三组和第四组接受双氯芬酸加水飞蓟素。结果显示,与对照组相比,第二组中过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GPx)和谷胱甘肽(GSH)的水平显著降低,而丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、碱性磷酸酶(ALP)、总胆红素、亚硝酸盐含量、丙二醛(MDA)、血清肿瘤坏死因子-α(TNF-α)和基因表达水平显著升高。另一方面,与第二组相比,水飞蓟素治疗导致CAT、SOD、GPx、GSH显著升高,MDA、ALT、AST、ALP、总胆红素、亚硝酸盐含量、血清TNF-α和基因表达显著降低。水飞蓟素给药也改善了组织病理学损伤。结果证实,水飞蓟素对双氯芬酸诱导的雄性大鼠肝毒性和氧化应激具有保护作用。
