A novel bacterial strain, designated KMB7, isolated from a freshwater pond in Taiwan, was characterized using a polyphasic taxonomic approach. Cells were Gram-stain-negative, motile by means of a single polar flagellum, rod-shaped and formed cream colonies. Optimal growth occurred at 25 °C, pH 7, and in the absence of NaCl. Phylogenetic analyses based on 16S rRNA gene sequences and an up-to-date bacterial core gene set (92 protein clusters) indicated that strain KMB7 is affiliated with species in the genus . The 16S rRNA gene sequence similarity indicated that strain KMB7 is closely related to species within the genus (95.2-97.6 % sequence similarity) and is most similar to CS-6 (97.6 %), followed by B6 (97.5 %). The average nucleotide identity and digital DNA-DNA hybridization identity between strain KMB7 and the closely related strains were 74.6-78.0 % and 19.0-21.2 %, respectively. The major fatty acids of strain KMB7 were summed feature 3 (C ω and/or C ω), C ω and C. The polar lipid profile consisted of a mixture of phosphatidylethanolamine, phosphatidylglycerol, phosphatidylserine, diphosphatidylglycerol and four unidentified phospholipids. The sole isoprenoid quinone was ubiquinone-8 (Q-8). Genomic DNA G+C content of strain KMB7 was 65.4 %. On the basis of phenotypic and genotypic properties and phylogenetic inference, strain KMB7 should be classified in a novel species of the genus , for which the name sp. nov. is proposed. The type strain is KMB7 (=BCRC 81156=LMG 30924=KCTC 62867).
一株从台湾淡水池塘中分离得到的新型细菌菌株 KMB7,通过多相分类学方法进行了特征描述。细胞革兰氏染色阴性,通过单一极生鞭毛运动,呈杆状,形成奶油色菌落。最佳生长条件为 25°C、pH 值 7 和无 NaCl。基于 16S rRNA 基因序列和最新的细菌核心基因集(92 个蛋白簇)的系统发育分析表明,菌株 KMB7 与属内的物种有关。16S rRNA 基因序列相似性表明,菌株 KMB7 与属内的种最为相似(95.2-97.6%序列相似性),与 CS-6(97.6%)最为相似,其次是 B6(97.5%)。菌株 KMB7 与密切相关菌株的平均核苷酸同一性和数字 DNA-DNA 杂交同一性分别为 74.6-78.0%和 19.0-21.2%。菌株 KMB7 的主要脂肪酸为 summed feature 3(C ω 和/或 C ω)、C ω 和 C。极性脂谱由磷脂酰乙醇胺、磷脂酰甘油、磷脂酰丝氨酸、双磷脂酰甘油和四种未鉴定的磷脂组成。唯一的异戊二烯醌是泛醌-8(Q-8)。菌株 KMB7 的基因组 DNA G+C 含量为 65.4%。基于表型和基因型特性以及系统发育推断,菌株 KMB7 应归类为属的一个新型种,建议命名为 sp. nov.。模式菌株为 KMB7(=BCRC 81156=LMG 30924=KCTC 62867)。
