To describe for the determination of contents of metabolites of benzene compounds in urine sample by high performance liquid chromatography. After acidification with hydrochloric acid, metabolites in urine were first extracted by acetonitrile and isopropanol (V∶V, 9∶1) with excessive sodium chloride, then gradient separated on a C18 column and then determined by DAD detector. There were good linear relationship between peak areas and injection quality in range of 2.00-100 mg/L (>0.999). The detection limit and quantitative limit of this method were 4.15-70.7 μg/L and 13.8-235 μg/L respectively. The precision for the analysis of urine was1.78%-8.23% ( =6). The average recovery of metabolites was 85.4%-105.5% at thee spiked levels in the range of 2.00-100 mg/L. The accuracy and reproducibility obtained make this method useful for the biological monitoring of occupational exposure to toluene, xylene, styrene and ethylbenzene.
描述用高效液相色谱法测定尿样中苯系物代谢产物的含量。尿样经盐酸酸化后,先用含过量氯化钠的乙腈和异丙醇(体积比9∶1)萃取其中的代谢产物,然后在C18柱上进行梯度分离,再用二极管阵列检测器(DAD)测定。在2.00 - 100 mg/L范围内峰面积与进样质量呈良好的线性关系(>0.999)。该方法的检出限和定量限分别为4.15 - 70.7 μg/L和13.8 - 235 μg/L。尿样分析的精密度为1.78% - 8.23%(n = 6)。在2.00 - 100 mg/L的加标水平下,代谢产物的平均回收率为85.4% - 105.5%。所获得的准确性和重现性使得该方法可用于甲苯、二甲苯、苯乙烯和乙苯职业接触的生物监测。
