Abstract

BACKGROUND

Numerous recent studies suggested that overexpression of the long noncoding RNA small nucleolar RNA host gene 12 (SNHG12) exhibited prooncogenic activity in multiple cancers. However, results regarding the prognostic value of SNHG12 in cancers still remained controversial. Therefore, we conducted a meta-analysis complemented with bioinformatics analysis to elucidate the clinical significance of SNHG12 in cancer patients.

METHODS

PubMed, Embase, Cochrane Library, Chinese National Knowledge Infrastructure, Wanfang, and Weipu databases were searched for eligible studies until July 2020. Additionally, bioinformatics analysis was applied to verify the results of meta-analysis.

RESULTS

Twenty-three related studies consisting of 1389 cancer patients were enrolled in the current meta-analysis. Elevated SNHG12 expression was found to be significantly associated with poor overall survival (OS) (HR = 1.81; 95% CI: 1.53-2.13; < 0.001) and disease-free survival (DFS) (HR = 1.40; 95% CI: 1.12-1.76; = 0.004) in multiple cancers, which were also verified by the results of bioinformatics analysis. Moreover, overexpression of SNHG12 was also related to clinicopathological characteristics including LNM, distant metastasis, high clinical stage, large tumor size, and poor tumor differentiation in diverse types of cancers.

CONCLUSION

The present findings indicated that SNHG12 might act as a novel biomarker for diagnosis or prognosis in human cancers.

大量近期研究表明,长链非编码 RNA 小核仁 RNA 宿主基因 12(SNHG12)的过表达在多种癌症中表现出致癌作用。然而,关于 SNHG12 在癌症中的预后价值的结果仍然存在争议。因此,我们进行了一项荟萃分析,并辅以生物信息学分析,以阐明 SNHG12 在癌症患者中的临床意义。

方法

检索 PubMed、Embase、Cochrane 图书馆、中国知网、万方和维普数据库,以获取合格的研究,截至 2020 年 7 月。此外,还应用生物信息学分析来验证荟萃分析的结果。

结果

本荟萃分析共纳入 23 项相关研究,包含 1389 名癌症患者。结果发现,SNHG12 表达升高与较差的总生存期(OS)(HR = 1.81;95% CI:1.53-2.13;<0.001)和无病生存期(DFS)(HR = 1.40;95% CI:1.12-1.76;= 0.004)显著相关,这一结果也通过生物信息学分析得到了验证。此外,SNHG12 的过表达还与包括淋巴结转移、远处转移、高临床分期、肿瘤较大和肿瘤分化不良在内的多种癌症的临床病理特征相关。

结论

本研究结果表明,SNHG12 可能作为人类癌症诊断或预后的新型生物标志物。
