A bacterial strain, designated 5J-6, was isolated from soil in Cheongnyeongpo, Republic of Korea. Cells were Gram-stain-positive, strictly aerobic, and motile rods and their catalase and oxidase activities were positive. Strain 5J-6 grew at 10-30 °C, pH 6.0-9.0, and 0-0.8% (w/v) NaCl concentration, with optimum growth at 25 °C, pH 6.5, and 0.4% NaCl concentration. Anteiso-C and iso-C were detected as the predominant fatty acids and menaquinone-7 was the sole isoprenoid quinone detected. Strain 5J-6 contained phosphatidylethanolamine, phosphatidylglycerol and an unidentified phospholipid as major polar lipids. The peptidoglycan belonged to the type A1γ meso-diaminopimelic acid. The G+C content of the genomic DNA calculated from the whole genomic sequence was 46.1 mol%. Phylogenetic analysis of strain 5J-6 based on 16S rRNA gene sequences placed the isolate into a member of the genus Paenibacillus. Sequence similarity analysis of 16S rRNA gene sequences revealed that strain 5J-6 was most closely related to Paenibacillus aceris KUDC4121 and Paenibacillus chondroitinus DSM 5051 with 98.76% and 98.42% similarities, respectively. Average nucleotide identity and in silico DNA-DNA hybridization values between strain 5J-6 and the type strain of P. aceris were 83.97% and 28.60%, respectively. Based on the phylogenetic and phenotypic characteristics and genomic data, strain 5J-6 could be considered to represent a novel species of the genus Paenibacillus, for which the name Paenibacillus silvestris sp. nov. is proposed. The type strain is 5J-6 (= KACC 21430 = JCM 33812).
从韩国淸宁浦土壤中分离出一株名为5J-6的细菌菌株。细胞革兰氏染色呈阳性,严格需氧,为能动杆菌,过氧化氢酶和氧化酶活性均为阳性。菌株5J-6在10-30°C、pH 6.0-9.0和0-0.8%(w/v)NaCl浓度下生长,最适生长温度为25°C,pH 6.5,NaCl浓度为0.4%。检测到anteiso-C和iso-C为主要脂肪酸,甲萘醌-7是唯一检测到的类异戊二烯醌。菌株5J-6含有磷脂酰乙醇胺、磷脂酰甘油和一种未鉴定的磷脂作为主要极性脂质。肽聚糖属于A1γ型中-二氨基庚二酸。根据全基因组序列计算,基因组DNA的G+C含量为46.1 mol%。基于16S rRNA基因序列对菌株5J-6进行系统发育分析,该分离株属于芽孢杆菌属。16S rRNA基因序列相似性分析表明,菌株5J-6与槭芽孢杆菌KUDC4121和软骨芽孢杆菌DSM 5051关系最为密切,相似性分别为98.76%和98.42%。菌株5J-6与槭芽孢杆菌模式菌株之间的平均核苷酸同一性和电子DNA-DNA杂交值分别为83.97%和28.60%。基于系统发育和表型特征以及基因组数据,菌株5J-6可被认为代表芽孢杆菌属的一个新物种,为此提出名为森林芽孢杆菌(Paenibacillus silvestris sp. nov.)。模式菌株为5J-6(=KACC 21430=JCM 33812)。
