Gastric cancer (GC) is one of the most common malignant tumors and the second most frequent cause of cancer death worldwide. Crocin is a kind of bioactive constituent found in the stigmas of saffron, which has shown various pharmacological activities.
In this study, we investigated the inhibitory effect of crocin on gastric cancer AGS cells proliferation and explored the underlying mechanism. A series of methods were used including cell counting kit assay, gene microarray analysis, qRT-PCR, Celigo image cytometry, cell clone formation assay, Western blot, and cell xenograft growth in vivo.
The results indicated that crocin inhibited AGS cells proliferation and promoted cell apoptosis. Further studies suggested that crocin decreased a series of genes expression, among which gene downregulation inhibited the tumor cells proliferation and tumor growth in mice, and overexpression of gene abolishes the inhibitory effect of crocin. Further study using microarray analysis suggested that knocking down of altered genes related to the proliferation and apoptosis of cells.
Crocin could inhibit the gastric cancer cells AGS cells proliferation by regulating gene expression, and may be a promising therapeutic target for GC treatment.
胃癌(GC)是最常见的恶性肿瘤之一,也是全球癌症死亡的第二大常见原因。藏红花素是一种在藏红花柱头中发现的生物活性成分,已显示出多种药理活性。
在本研究中,我们研究了藏红花素对胃癌AGS细胞增殖的抑制作用,并探讨了其潜在机制。使用了一系列方法,包括细胞计数试剂盒检测、基因芯片分析、qRT-PCR、Celigo图像细胞仪检测、细胞克隆形成试验、蛋白质免疫印迹法以及体内细胞异种移植生长实验。
结果表明,藏红花素抑制AGS细胞增殖并促进细胞凋亡。进一步研究表明,藏红花素降低了一系列基因的表达,其中基因下调抑制了小鼠肿瘤细胞的增殖和肿瘤生长,而该基因的过表达消除了藏红花素的抑制作用。使用芯片分析的进一步研究表明,敲低改变的基因与细胞的增殖和凋亡有关。
藏红花素可通过调节基因表达抑制胃癌AGS细胞增殖,并且可能是胃癌治疗的一个有前景的治疗靶点。
