Abstract

BACKGROUND

In myocardial infarction (MI), inflammatory cytokine including tumor necrosis factor-α (TNF-α) plays pivotal role throughout worsening processes and recovery, whereas its cleavage is controlled by a disintegrin and metalloproteinases-17 (ADAM-17). (Wight) is widely used plant in Indonesia and Southeast Asian countries to treat various illnesses.

OBJECTIVE

This study aimed to analyze the effect of extract towards TNF-α and ADAM17 expressions in MI rat model.

METHODS

Myocardial infarction were surgically induced in wistar rats by ligating left anterior descending coronary artery in both treatment and control group. Around 3.6 mg of extract (SPE) was administered via nasogastric tube to treatment groups, while control group received only saline. Serum TNF-α level and expression of TNF-α and ADAM17 in blanching heart muscle was examined in both groups on day 1, day 4, day 7, and day 14 after treatment with SPE.

RESULTS

Reduction of serum TNF-α levels was markedly seen from day 4 in treatment group and was lower than in control group since day 4. Expression of ADAM17 was reduced and expression of TNF-α was only seen in myocardial membrane in group with SPE treatment.

CONCLUSION

exerts its anti-inflammatory effect by decreasing ADAM17 expression subsequently lowering TNF-α regulation in MI rat model.

背景

在心肌梗死(MI)中,炎症细胞因子,包括肿瘤坏死因子-α(TNF-α),在病情恶化和恢复过程中发挥着关键作用,而其裂解受解整合素金属蛋白酶 17(ADAM-17)的控制。(Wight)在印度尼西亚和东南亚国家广泛用作治疗各种疾病的植物。

目的

本研究旨在分析 提取物对 MI 大鼠模型中 TNF-α和 ADAM17 表达的影响。

方法

通过结扎左前降支冠状动脉在治疗组和对照组大鼠中诱导心肌梗死。治疗组通过鼻胃管给予约 3.6 毫克 提取物(SPE),对照组仅给予生理盐水。在 SPE 治疗后第 1、4、7 和 14 天,分别检测两组大鼠血清 TNF-α水平以及心肌组织中 TNF-α和 ADAM17 的表达。

结果

治疗组从第 4 天开始,血清 TNF-α水平明显下降,且自第 4 天起低于对照组。ADAM17 的表达减少,仅在 SPE 治疗组的心肌膜中观察到 TNF-α的表达。

结论

在 MI 大鼠模型中, 通过降低 ADAM17 的表达,进而降低 TNF-α的调节,发挥其抗炎作用。
