Abstract

OBJECTIVE

It remains unclear whether human T-cell leukemia virus type 1 (HTLV-1) infection influences therapeutic responses in patients with rheumatic diseases and whether immunosuppressive treatments increase the risk of HTLV-1-related complications in HTLV-1 carriers with rheumatic diseases. We examined the effects of tocilizumab (TCZ), an interleukin (IL)-6 receptor antagonist, on two HTLV-1-infected T-cell lines (HCT-5 and MT-2) .

METHODS

We evaluated production of cytokines and chemokines, expression of HTLV-I associated genes, HTLV-1 proviral load (PVL), expression of HTLV-1 structural proteins, and apoptosis.

RESULTS

There were no significant differences in cytokine and chemokine levels in the culture supernatants of HCT-5 and MT-2 cells treated with phosphate-buffered saline (PBS) or TCZ. No significant differences were detected in mRNA abundance of Tax or HBZ, PVL, expression of the HTLV-1 structural protein GAG, or apoptosis among HCT-5 and MT-2 cells treated with PBS or TCZ.

CONCLUSIONS

TCZ had no effect the cytokine profiles, HTLV-1 gene and protein expression, PVL, or apoptosis in HTLV-1-infected T-cell lines. Thus, TCZ treatment has no effect on HTLV-1 infection .

目的

目前尚不清楚人类1型T细胞白血病病毒(HTLV-1)感染是否会影响风湿性疾病患者的治疗反应,以及免疫抑制治疗是否会增加患有风湿性疾病的HTLV-1携带者发生HTLV-1相关并发症的风险。我们研究了白细胞介素(IL)-6受体拮抗剂托珠单抗(TCZ)对两种HTLV-1感染的T细胞系(HCT-5和MT-2)的影响。

方法

我们评估了细胞因子和趋化因子的产生、HTLV-I相关基因的表达、HTLV-1前病毒载量(PVL)、HTLV-1结构蛋白的表达以及细胞凋亡情况。

结果

用磷酸盐缓冲盐水(PBS)或TCZ处理的HCT-5和MT-2细胞培养上清液中的细胞因子和趋化因子水平没有显著差异。在用PBS或TCZ处理的HCT-5和MT-2细胞中,Tax或HBZ的mRNA丰度、PVL、HTLV-1结构蛋白GAG的表达或细胞凋亡均未检测到显著差异。

结论

TCZ对HTLV-1感染的T细胞系中的细胞因子谱、HTLV-1基因和蛋白表达、PVL或细胞凋亡没有影响。因此,TCZ治疗对HTLV-1感染没有作用。
