Abstract

BACKGROUND

as an important parasite affects health of humans and animals in some tropical and subtropical areas of the world, including Iran. Little is known about the molecular diversity of in Equidae. Therefore, this study aimed to characterize the genetic polymorphisms among parasites.

METHODS

Eight adult spp. isolates were collected from a working donkey after necropsy in Shiraz, southwestern Iran, in 2018. Primarily, various parameters were measured morphologically. Subsequently, DNA was extracted from each fluke and molecular markers of cytochrome C oxidase (1) and NADH dehydrogenase 1(1) from individual isolates were amplified using PCR assay and sequence data were employed for molecular and phylogenetic analysis. Genetic diversity between isolates was evaluated by comparing the sequences of these two mitochondrial regions.

RESULTS

Based on the morphological and analyzed mitochondrial sequences, all of eight donkey isolates (100%) were identified as . Moreover, nine and five nucleotide polymorphisms were identified in the 1and 1 region sequences, respectively.

CONCLUSION

Accordingly, phylogenetic data revealed five and four haplotypes among donkey isolates based on the 1and 1 markers. Similarly, some of these haplotypes have been previously reported from different host species in Iran as well as all around the world.

背景

作为一种重要的寄生虫,在世界上一些热带和亚热带地区(包括伊朗)影响着人类和动物的健康。关于马科动物体内该寄生虫的分子多样性知之甚少。因此,本研究旨在对该寄生虫的遗传多态性进行特征分析。

方法

2018年在伊朗西南部设拉子对一头工作用驴进行尸检后,收集了8个成年该寄生虫分离株。首先,对各种参数进行形态学测量。随后,从每个吸虫中提取DNA,使用PCR检测对各个分离株的细胞色素C氧化酶1(COX1)和烟酰胺腺嘌呤二核苷酸脱氢酶1(ND1)分子标记进行扩增,并将序列数据用于分子和系统发育分析。通过比较这两个线粒体区域的序列来评估分离株之间的遗传多样性。

结果

根据形态学和分析的线粒体序列,所有8个驴分离株(100%)均被鉴定为该寄生虫。此外,在COX1和ND1区域序列中分别鉴定出9个和5个核苷酸多态性。

结论

因此,系统发育数据显示基于COX1和ND1标记,驴分离株中有5个和4个单倍型。同样,这些单倍型中的一些先前已在伊朗以及世界各地的不同宿主物种中报道过。
