We aimed to explore the diagnostic values of miR-221-3p in serum and cerebrospinal fluid (CSF) for post-stroke depression (PSD) and to analyze the risk factors of the disease.
Admitted to the Second Affiliated Hospital of Harbin Medical University, Harbin, China from May 2013 to May 2020, 136 stroke patients were enrolled, among which 76 PSD patients were taken as a PSD group and 60 non-depressed patients were taken as a Non-PSD group. miR-221-3p expression in serum and CSF and concentrations of inflammatory cytokines (IL-6, TNF-α) in serum were detected, to analyze the diagnostic and prognostic values of the indicators for PSD. Correlations of miR-221-3p in serum with that in CSF, with the National Institute of Health Stroke Scale (NIHSS) score and the Hamilton Depression Rating Scale (HAMD) score, and with inflammatory cytokines were analyzed, so as to analyze the risk factors affecting the occurrence of PSD.
Compared with the Non-PSD group, miR-221-3p remarkably upregulated in serum and CSF in the PSD group, and its areas under the curves (AUCs) for PSD identification were 0.900 and 0.925, respectively. According to the correlation analysis, miR-221-3p in serum was remarkably positively correlated with that in CSF, NIHSS score, HAMD score, IL-6 and TNF-α. In addition, a history of mental illness, NIHSS score, HAMD score, IL-6, TNF-α and miR-221-3p were risk factors of PSD.
miR-221-3p in serum and CSF can be used as the diagnostic and risk warning indicators of PSD.
我们旨在探讨血清和脑脊液(CSF)中miR-221-3p对卒中后抑郁(PSD)的诊断价值,并分析该疾病的危险因素。
选取2013年5月至2020年5月在中国哈尔滨医科大学附属第二医院收治的136例卒中患者,其中76例PSD患者作为PSD组,60例非抑郁患者作为非PSD组。检测血清和脑脊液中miR-221-3p的表达以及血清中炎性细胞因子(IL-6、TNF-α)的浓度,以分析这些指标对PSD的诊断和预后价值。分析血清中miR-221-3p与脑脊液中miR-221-3p、与美国国立卫生研究院卒中量表(NIHSS)评分和汉密尔顿抑郁量表(HAMD)评分以及与炎性细胞因子的相关性,以分析影响PSD发生的危险因素。
与非PSD组相比,PSD组血清和脑脊液中miR-221-3p显著上调,其用于PSD识别的曲线下面积(AUC)分别为0.900和0.925。根据相关性分析,血清中miR-221-3p与脑脊液中miR-221-3p、NIHSS评分、HAMD评分、IL-6和TNF-α显著正相关。此外,精神病史、NIHSS评分、HAMD评分、IL-6、TNF-α和miR-221-3p是PSD的危险因素。
血清和脑脊液中的miR-221-3p可作为PSD的诊断和风险预警指标。
