A beige-pigmented, Gram-strain-negative, aerobic, rod-shaped, non-flagellated and non-gliding bacterium, designated strain lm94, was isolated from rhizosphere soil of Alhagi sparsifolia obtained from Alar city, located in Xinjiang province, China. Growth occurred at 20-45 °C (optimum, 37 °C), in the presence of 0-6% (w/v) NaCl (optimum, 0-1%) and at pH 6.0-9.5 (optimum, pH 7.0-7.5). Phylogenetic analysis based on 16S rRNA gene sequence showed that strain lm94 belonged to the genus Mesorhizobium, with highest sequence similarity to Mesorhizobium wenxiniae WYCCWR 10195 (96.6%). Genome sequencing revealed a genome size of 5 256 375 bp and a G + C content of 63.6 mol%. The average nucleotide identity value and the digital DNA-DNA hybridization value between strain lm94 and M. wenxiniae LMG 30254 were 75.0% and 20.0%, respectively. The major respiratory quinone was Q-10. The major fatty acids were C cyclo ω8c and Summed Feature 8 (C ω6c and/or C ω7c) and its polar lipids consisted of phosphatidylethanolamine (PE), phosphatidylglycerol (PG), unidentified phospholipid (PL), phosphatidylcholine (PC), diphosphatidylglycerol (DPG), unidentified aminolipid (AL), unknown glycolipid (GL), unidentified aminophospholipid (APL2) and unidentified polar lipid (L1 and L2). On the basis of these data, strain lm94 is considered to represent a novel species of the genus Mesorhizobium, for which the name Mesorhizobium xinjiangense sp. nov. is proposed. The type strain is lm94 (=KCTC 72863=CCTCC AB2019377).
从中国新疆阿拉尔市的疏叶骆驼刺根际土壤中分离出一株米色色素、革兰氏染色阴性、需氧、杆状、无鞭毛且不滑动的细菌,命名为菌株lm94。该菌株在20-45°C(最适温度为37°C)、0-6%(w/v)NaCl(最适浓度为0-1%)以及pH值为6.0-9.5(最适pH值为7.0-7.5)的条件下生长。基于16S rRNA基因序列的系统发育分析表明,菌株lm94属于中生根瘤菌属,与文新中生根瘤菌WYCCWR 10195的序列相似性最高(96.6%)。基因组测序显示,其基因组大小为5256375 bp,G+C含量为63.6 mol%。菌株lm94与文新中生根瘤菌LMG 30254之间的平均核苷酸同一性值和数字DNA-DNA杂交值分别为75.0%和20.0%。主要呼吸醌为Q-10。主要脂肪酸为C环ω8c和总和特征8(C ω6c和/或C ω7c),其极性脂质由磷脂酰乙醇胺(PE)、磷脂酰甘油(PG)、未鉴定的磷脂(PL)、磷脂酰胆碱(PC)、二磷脂酰甘油(DPG)、未鉴定的氨基脂质(AL)、未知糖脂(GL)、未鉴定的氨基磷脂(APL2)以及未鉴定的极性脂质(L1和L2)组成。基于这些数据,菌株lm94被认为代表了中生根瘤菌属的一个新物种,为此提出新名称新疆中生根瘤菌(Mesorhizobium xinjiangense sp. nov.)。模式菌株为lm94(=KCTC 72863=CCTCC AB2019377)。
