Abstract

BACKGROUND

The purpose of the present study was to investigate the antimicrobial effects of berberine and thioridazine, as well as their effect on the gene expression of the AdeABC efflux pump system in Multidrug-Resistant (MDR) () isolates.

METHODS

This study was carried out in five MDR clinical isolates of and a sample of standard strain ( PTCC1797). The effect of Minimum Inhibitory Concentration (MIC) and the Minimum Bactericidal Concentration (MBC) of berberine, thioridazine, and ciprofloxacin alone and their combination on was evaluated by broth microdilution method. Also, their effect on the expression of efflux pump gene was evaluated using real time PCR method.

RESULTS

The MIC of thioridazine, berberine, ciprofloxacin+thioridazine, ciprofloxacin+ berberine, thioridazine+berberine, and ciprofloxacin+thioridazine+berberine on MDR A. baumannii isolates was 64, 256, 128, 256, 128, and 128 μg/ml, respectively. The results showed that treatment of strains with thioridazine alone and in combination with berberine and ciprofloxacin significantly (p<0.05) decreased the expression of efflux pump gene.

CONCLUSION

Due to the inhibitory effects of thioridazine on bacterial isolates and efflux pump gene, this compound can be used as a potential antimicrobial agent against MDR A.

背景

本研究旨在探讨黄连素和硫利达嗪的抗菌作用,以及它们对多重耐药(MDR)鲍曼不动杆菌分离株中AdeABC外排泵系统基因表达的影响。

方法

本研究对五株MDR鲍曼不动杆菌临床分离株和一株标准菌株(ATCC 1797)进行了研究。采用肉汤微量稀释法评估了黄连素、硫利达嗪、环丙沙星单独及联合使用时的最低抑菌浓度(MIC)和最低杀菌浓度(MBC)对鲍曼不动杆菌的影响。此外,使用实时PCR法评估了它们对AdeABC外排泵基因表达的影响。

结果

硫利达嗪、黄连素、环丙沙星+硫利达嗪、环丙沙星+黄连素、硫利达嗪+黄连素以及环丙沙星+硫利达嗪+黄连素对MDR鲍曼不动杆菌分离株的MIC分别为64、256、128、256、128和128μg/ml。结果表明,单独使用硫利达嗪以及将其与黄连素和环丙沙星联合使用处理菌株时,显著(p<0.05)降低了AdeABC外排泵基因的表达。

结论

由于硫利达嗪对细菌分离株和AdeABC外排泵基因具有抑制作用,该化合物可作为一种潜在的抗MDR鲍曼不动杆菌的抗菌剂。
