Abstract

The purpose of this study was the production of maltobionic acid, in the form of sodium maltobionate, by Z. mobilis cells immobilized in polyurethane. The in situ immobilized system (0.125-0.35 mm) was composed of 7 g polyol, 3.5 g isocyanate, 0.02 g silicone, and 7 g Z. mobilis cell, at the concentration of 210 g/L. The bioconversion of maltose to sodium maltobionate was performed with different cell concentrations (7.0-9.0 g/L), temperature (30.54-47.46 °C), pH (5.55-7.25), and substrate concentration (0.7-1.3 mol/L). The stability of the immobilized system was evaluated for 24 h bioconversion cycles and storage of 6 months. The maximum concentration of sodium maltobionate was 648.61 mmol/L in 34.34 h process (8.5 g/L) at 39 °C and pH 6.30. The immobilized system showed stability for 19 successive operational cycles of 24 h bioconversion and 6 months of storage, at 4 °C or 22 °C.

本研究的目的是利用固定在聚氨酯中的运动发酵单胞菌细胞生产麦芽酮酸,以麦芽酮酸钠的形式存在。原位固定化系统(0.125-0.35 毫米)由 7 克多元醇、3.5 克异氰酸酯、0.02 克硅酮和 7 克运动发酵单胞菌细胞组成,浓度为 210 克/升。在不同细胞浓度(7.0-9.0 克/升)、温度(30.54-47.46°C)、pH(5.55-7.25)和底物浓度(0.7-1.3 摩尔/升)下,进行麦芽糖向麦芽酮酸钠的生物转化。评价了固定化系统在 24 小时生物转化循环和 6 个月储存期间的稳定性。在 39°C 和 pH 6.30 下,经过 34.34 小时的过程(8.5 克/升),最大的麦芽酮酸钠浓度为 648.61 毫摩尔/升。固定化系统在 4°C 或 22°C 下,经过 19 次连续 24 小时生物转化操作循环和 6 个月的储存,表现出稳定性。
