To investigate the therapeutic potential of nano-emulsion of mangosteen rind extract in a mucoadhesive gingival patch on periodontitis, and its effect on tumor necrosis factor alpha (TNF-α), receptor activator of nuclear factor kappa Β ligand (RANKL), and interleukin 10 (IL-10) expression.
Sixty Wistar rats were divided into four groups: positive control group (mucoadhesive patch with doxycycline), negative control group (mucoadhesive patch), treatment group I (mucoadhesive patch with mangosteen rind extract), and treatment group II (mucoadhesive patch with nano-emulsion of mangosteen rind extract). An experimental model of -induced periodontitis was established in rats by treatment with 0.03 mL bacteria locally (1 × 10 colony-forming units) seven times at 2-day intervals in the gingival sulcus of mandibular anterior teeth. Treatment was 1 h/day for 3 days. On days 3, 5, and 7, five rats from each group were killed. TNF-α, IL-10, and RANKL expression was determined by dissecting the lower jaw for immunohistochemistry.
The mucoadhesive patch with nano-emulsion mangosteen rind extract significantly decreased TNF-α and RANKL expression and increased IL-10 expression (p < 0.05) compared to the treatment I, positive and negative control groups.
A mucoadhesive gingival patch with nano-emulsion of mangosteen rind extract has the potential to treat periodontitis by decreasing TNF-α, RANKL, and increasing IL-10 expression.
研究山竹果皮提取物纳米乳剂在黏附性牙龈贴片中对牙周炎的治疗潜力,及其对肿瘤坏死因子α(TNF-α)、核因子κB受体激活剂配体(RANKL)和白细胞介素10(IL-10)表达的影响。
将60只Wistar大鼠分为四组:阳性对照组(含强力霉素的黏附贴片)、阴性对照组(黏附贴片)、治疗组I(含山竹果皮提取物的黏附贴片)和治疗组II(含山竹果皮提取物纳米乳剂的黏附贴片)。通过在下颌前牙牙龈沟内每隔2天局部注射0.03 mL细菌(1×10菌落形成单位)7次,建立大鼠诱导性牙周炎实验模型。治疗为期3天,每天1小时。在第3、5和7天,每组处死5只大鼠。通过解剖下颌进行免疫组织化学测定TNF-α、IL-10和RANKL的表达。
与治疗组I、阳性和阴性对照组相比,含山竹果皮提取物纳米乳剂的黏附贴片显著降低了TNF-α和RANKL的表达,并增加了IL-10的表达(p<0.05)。
含山竹果皮提取物纳米乳剂的黏附性牙龈贴片有可能通过降低TNF-α、RANKL并增加IL-10的表达来治疗牙周炎。
