Abstract

Bioactive polysaccharides have numerous pharmacological effects that are beneficial to human health. Akebia trifoliata (Thunb.) Koidz. has great development prospects as a food resource with medicinal value. The polysaccharides (ATFP) were extracted from A. trifoliata fruit by an aqueous two-phase system. ATFP-3, purified with DEAE-52 and Sephadex G-200 from ATFP, was mainly composed of glucose (47.55%) and galactose (20.39%). Its hydroxyl radical scavenging rate was 89.30% at 1.60 mg/mL and its IC was 0.29 mg/mL. ATFP-3 significantly enhanced the survival rate of Caenorhabditis elegans under thermal or oxidative stress. Furthermore, ATFP-3 could prolong the lifespan of C. elegans and improve the activities of the antioxidant enzyme, while also decrease the accumulation of lipofuscin and the level of malondialdehyde (MDA) in aging worms. Thus, ATFP-3 has application potential in health benefits for humans.

生物活性多糖具有许多对人体健康有益的药理作用。三叶木通作为一种具有药用价值的食物资源具有很大的开发前景。采用双水相体系从三叶木通果实中提取多糖(ATFP)。用DEAE-52和Sephadex G-200从ATFP中纯化得到的ATFP-3主要由葡萄糖(47.55%)和半乳糖(20.39%)组成。其在1.60mg/mL时的羟自由基清除率为89.30%,IC50为0.29mg/mL。ATFP-3显著提高了秀丽隐杆线虫在热应激或氧化应激下的存活率。此外,ATFP-3可以延长秀丽隐杆线虫的寿命,提高抗氧化酶的活性,同时还能减少衰老线虫中脂褐素的积累和丙二醛(MDA)的水平。因此,ATFP-3在对人类健康有益方面具有应用潜力。
