This study aimed to evaluate the effects of maternal nucleotide (NT) supplementation on intestinal morphology and immune function in lipopolysaccharide-challenged newborn piglets. At 85 d gestation, 12 sows were selected and assigned to two groups: the CON group (basal diet, = 6) and the NT group (basal diet with 1 g/kg NT mixture, = 6). After parturition, newborn piglets were collected without suckling. Piglets from the CON group were intraperitoneally injected with sterile saline or lipopolysaccharide (LPS, 10 mg/kg body weight), and divided into the C-CON ( = 6) and C-LPS groups ( = 6). Piglets from the NT group received the same treatment and were divided into the N-CON ( = 6) and N-LPS groups ( = 6). The blood and small intestinal samples of piglets were collected 1 h after injection. The results showed that: (1) maternal NT supplementation increased the concentrations of serum complement C3 and C4 ( < 0.05), and suppressed the increase in serum hypersensitive C-reactive protein in LPS-challenged newborn piglets ( < 0.05); (2) maternal NT supplementation increased the villus height and the ratio of villus height to crypt depth in the duodenum of newborn piglets ( < 0.05) and inhibited the LPS-induced decrease in the villus height in the jejunum and ileum ( < 0.05). (3) The LPS-induced increased levels of interleukin-6 in the jejunum and tumor necrosis factor-α in the ileum of newborn piglets were suppressed by maternal NT supplementation ( < 0.05). (4) In the jejunum of newborn piglets, maternal NT supplementation inhibited the LPS-induced increase in toll-like receptor 4 (TLR4) mRNA and protein expression ( < 0.05) and the decrease of nuclear factor-κB inhibitor α (IκBα) protein expression ( < 0.05). In the ileum, piglets had a lower nuclear factor-κB (NFκB) mRNA expression in the NT groups than the CON groups ( < 0.05), and maternal NT supplementation suppressed the decrease of IκBα mRNA in LPS-treated piglets ( < 0.05). In conclusion, maternal NT supplementation could promote the intestinal development and immune function of newborn piglets, and may improve LPS-induced intestinal inflammatory responses the TLR4/IκBα/NFκB pathway.
本研究旨在评估母体补充核苷酸(NT)对脂多糖刺激的新生仔猪肠道形态和免疫功能的影响。在妊娠85天时,选择12头母猪并分为两组:CON组(基础日粮,n = 6)和NT组(基础日粮+1 g/kg NT混合物,n = 6)。分娩后,收集未哺乳的新生仔猪。CON组仔猪腹腔注射无菌生理盐水或脂多糖(LPS,10 mg/kg体重),分为C-CON组(n = 6)和C-LPS组(n = 6)。NT组仔猪接受相同处理,分为N-CON组(n = 6)和N-LPS组(n = 6)。注射后1小时收集仔猪的血液和小肠样本。结果表明:(1)母体补充NT可提高脂多糖刺激的新生仔猪血清补体C3和C4浓度(P<0.05),并抑制血清超敏C反应蛋白的升高(P<0.05);(2)母体补充NT可增加新生仔猪十二指肠绒毛高度和绒毛高度与隐窝深度比值(P<0.05),并抑制LPS诱导的空肠和回肠绒毛高度降低(P<0.05)。(3)母体补充NT可抑制LPS诱导的新生仔猪空肠白细胞介素-6水平升高和回肠肿瘤坏死因子-α水平升高(P<0.05)。(4)在新生仔猪空肠中,母体补充NT可抑制LPS诱导的Toll样受体4(TLR4)mRNA和蛋白表达增加(P<0.05)以及核因子-κB抑制因子α(IκBα)蛋白表达降低(P<0.05)。在回肠中,NT组仔猪核因子-κB(NFκB)mRNA表达低于CON组(P<0.05),母体补充NT可抑制LPS处理仔猪IκBα mRNA的降低(P<0.05)。总之,母体补充NT可促进新生仔猪肠道发育和免疫功能,并可能通过TLR4/IκBα/NFκB途径改善LPS诱导的肠道炎症反应。
