A Gram-staining-positive, non-motile, aerobic, spherical actinobacterium, designated WL0053, was isolated from the coastal sediment of Nantong City, Jiangsu Province, China. The 16S rRNA gene sequence of strain WL0053 exhibited the highest similarities to Nocardioides mesophilus MSL-22 (98.0%), N. massiliensis GD12 (97.8%), Marmoricola bigeumensis MSL-05 (97.6%), and N. jensenii DSM 20641 (97.3%). The polyphasic taxonomic approach was used for the identification of strain WL0053. This strain formed white, round, and smooth colonies and grew in the presence of 0-18% (w/v) NaCl (optimum, 0-4.0%), at pH 6.0-9.0 (optimum, pH 7.0) and at 20-37 °C (optimum, 28 °C). The main cellular fatty acids comprised of C ω8c, C ω9c, and iso-C. The genomic DNA G + C content was 71.9%. The predominant quinone was MK-8(H), and the major polar lipid consisted of phosphatidylcholine, glycolipid, phosphatidylethanolamine, and two unidentified phospholipids. Phylogenetic trees of 16S rRNA gene and bac120 gene set indicted that strain WL0053 was closely related to the species N. iriomotensis and N. mesophilus, while these two species clustered in a separate clade together with M. caldifontis YIM 730233 in the bac120 tree. Combined with the analysis of average nucleotide identity (ANI), average amino acid identity (AAI), and digital DNA-DNA hybridization (dDDH), it can be considered that the strain WL0053 is a new member of the genus Nocardioides and is proposed to be named as Nocardioides jiangsuensis sp. nov.. The type strain is WL0053 (=MCCC 1K05897=JCM 34671=GDMCC 4.192). Furthermore, based on the fact that the genera Nocardioides and Marmoricola both appeared polyphyletic with no significant difference on phenotypic and chemotaxonomic traits, we proposed to reclassify the genus Marmoricola as Nocardioides.
从中国江苏省南通市沿海沉积物中分离出一株革兰氏染色阳性、不运动、需氧的球形放线菌,命名为WL0053。菌株WL0053的16S rRNA基因序列与嗜中诺卡氏菌MSL-22(98.0%)、马赛诺卡氏菌GD12(97.8%)、大隅海栖孢菌MSL-05(97.6%)和詹氏诺卡氏菌DSM 20641(97.3%)的相似性最高。采用多相分类方法对菌株WL0053进行鉴定。该菌株形成白色、圆形、光滑的菌落,在0-18%(w/v)NaCl(最适0-4.0%)、pH 6.0-9.0(最适pH 7.0)和20-37℃(最适28℃)条件下生长。主要细胞脂肪酸包括C ω8c、C ω9c和异-C。基因组DNA G+C含量为71.9%。主要醌为MK-8(H),主要极性脂由磷脂酰胆碱、糖脂、磷脂酰乙醇胺和两种未鉴定的磷脂组成。16S rRNA基因和bac120基因集的系统发育树表明,菌株WL0053与冲绳诺卡氏菌和嗜中诺卡氏菌关系密切,而在bac120树中,这两个物种与嗜热栖泉菌YIM 730233聚集在一个单独的分支中。结合平均核苷酸同一性(ANI)、平均氨基酸同一性(AAI)和数字DNA-DNA杂交(dDDH)分析,可认为菌株WL0053是诺卡氏菌属的一个新成员,建议命名为江苏诺卡氏菌新种。模式菌株为WL0053(=MCCC 1K05897=JCM 34671=GDMCC 4.192)。此外,基于诺卡氏菌属和海栖孢菌属在表型和化学分类特征上均呈现多系性且无显著差异这一事实,我们建议将海栖孢菌属重新分类为诺卡氏菌属。
