To investigate the relationship between serum long non-coding RNA (lncRNA) X inactive specific transcript (XIST) and microRNA-30d-5p (miR-30d-5p) expression levels in type 2 diabetic peripheral neuropathy (DPN).
Clinical data of patients with only type 2 diabetes mellitus (pure T2DM group), DPN patients (DPN group) and healthy patients (control group) admitted to Inner Mongolia Forestry General Hospital from August 2019 to April 2022 were retrospectively analyzed, with 76 cases in each group. The serum lncRNA XIST and miR-30d-5p expression levels of each group were compared. The correlation between serum lncRNA XIST and miR-30d-5p in DPN patients was analyzed. The influencing factors of DPN occurrence were analyzed. Also, the diagnostic value of serum lncRNA XIST and miR-30d-5p for DPN was analyzed.
There were significant differences in the lncRNA XIST and miR-30d-5p expression levels among the pure T2DM group, DPN group, and control group. LncRNA XIST expression level was negatively correlated with miR-30d-5p in DPN patients (<0.05). Triglycerides, hemoglobin A1c, miR-30d-5p were risk factors for the occurrence of DPN, and lncRNA XIST was a protective factor (<0.05). The areas under the curve (AUC) of serum lncRNA XIST and miR-30d-5p for the diagnosis of DPN were 0.851 and 0.845, respectively, and the AUC of lncRNA XIST and miR-30d-5p combined was 0.932, with a sensitivity of 92.1%, and a specificity of 85.5%.
Both lncRNA XIST and miR-30d-5p may be involved in the development of type 2 DPN. Therefore, detecting serum levels of both may be helpful for clinical diagnosis and treatment of type 2 DPN.
探讨2型糖尿病周围神经病变(DPN)患者血清长链非编码RNA(lncRNA)X染色体失活特异性转录本(XIST)与微小RNA-30d-5p(miR-30d-5p)表达水平之间的关系。
回顾性分析2019年8月至2022年4月内蒙古林业总医院收治的单纯2型糖尿病患者(单纯T2DM组)、DPN患者(DPN组)和健康患者(对照组)的临床资料,每组76例。比较各组血清lncRNA XIST和miR-30d-5p的表达水平。分析DPN患者血清lncRNA XIST与miR-30d-5p的相关性。分析DPN发生的影响因素。同时,分析血清lncRNA XIST和miR-30d-5p对DPN的诊断价值。
单纯T2DM组、DPN组和对照组的lncRNA XIST和miR-30d-5p表达水平存在显著差异。DPN患者中lncRNA XIST表达水平与miR-30d-5p呈负相关(<0.05)。甘油三酯、糖化血红蛋白A1c、miR-30d-5p是DPN发生 的危险因素,lncRNA XIST是保护因素(<0.05)。血清lncRNA XIST和miR-30d-5p诊断DPN的曲线下面积(AUC)分别为0.851和0.845,lncRNA XIST与miR-30d-5p联合检测的AUC为0.932,灵敏度为92.1%,特异度为85.5%。
lncRNA XIST和miR-30d-5p可能均参与2型DPN的发生发展。因此,检测二者的血清水平可能有助于2型DPN的临床诊断和治疗。
