Two heterotrophic bacteroidetes strains were isolated as satellites from autotrophic enrichments inoculated with samples from hypersaline soda lakes in southwestern Siberia. Strain Z-1702 is an obligate anaerobic fermentative saccharolytic bacterium from an iron-reducing enrichment culture, while Ca. Cyclonatronum proteinivorum Omega is an obligate aerobic proteolytic microorganism from a cyanobacterial enrichment. Cells of isolated bacteria are characterized by highly variable morphology. Both strains are chloride-independent moderate salt-tolerant obligate alkaliphiles and mesophiles. Strain Z-1702 ferments glucose, maltose, fructose, mannose, sorbose, galactose, cellobiose, N-acetyl-glucosamine and alpha-glucans, including starch, glycogen, dextrin, and pullulan. Strain Omega is strictly proteolytic utilizing a range of proteins and peptones. The main polar lipid fatty acid in both strains is iso-C, while other major components are various C and C isomers. According to pairwise sequence alignments using BLAST Gracilimonas was the nearest cultured relative to both strains (<90% of 16S rRNA gene sequence identity). Phylogenetic analysis placed strain Z-1702 and strain Omega as two different genera in a deep-branching clade of the new family level within the order Balneolales with genus. Based on physiological characteristics and phylogenetic position of strain Z-1702 it was proposed to represent a novel genus and species Natronogracilivirga saccharolityca gen. nov., sp. nov. (= DSMZ 109061 =JCM 32930 =VKM B 3262). Furthermore, phylogenetic and phenotypic parameters of N. saccharolityca and C. proteinivorum gen. nov., sp. nov., strain Omega (=JCM 31662, =UNIQEM U979), make it possible to include them into a new family with a proposed designation Cyclonatronaceae fam. nov..
从西伯利亚西南部高盐苏打湖样品接种的自养富集培养物中分离出两株异养拟杆菌菌株作为卫星菌。菌株Z-1702是一株来自铁还原富集培养物的专性厌氧发酵性糖分解细菌,而类环钠嗜盐菌Omega是一株来自蓝细菌富集培养物的专性需氧蛋白水解微生物。分离出的细菌细胞具有高度可变的形态。两株菌株均为不依赖氯化物的中度耐盐专性嗜碱菌和嗜温菌。菌株Z-1702发酵葡萄糖、麦芽糖、果糖、甘露糖、山梨糖、半乳糖、纤维二糖、N-乙酰葡糖胺和α-葡聚糖,包括淀粉、糖原、糊精和支链淀粉。菌株Omega严格进行蛋白水解,利用多种蛋白质和蛋白胨。两株菌株中的主要极性脂质脂肪酸均为异-C,而其他主要成分是各种C和C异构体。根据使用BLAST进行的成对序列比对,纤细单胞菌属是与两株菌株亲缘关系最近的已培养菌属(16S rRNA基因序列同一性<90%)。系统发育分析将菌株Z-1702和菌株Omega置于盐浴菌目内新科水平的一个深分支进化枝中的两个不同属。基于菌株Z-1702的生理特征和系统发育位置,提议将其代表一个新的属和种,即嗜盐纤细弧菌属糖分解嗜盐纤细弧菌(Natronogracilivirga saccharolityca),新属,新种(=DSMZ 109061 =JCM 32930 =VKM B 3262)。此外,嗜盐糖分解嗜盐纤细弧菌和类环钠嗜盐菌新属,新种,菌株Omega(=JCM 31662,=UNIQEM U979)的系统发育和表型参数,使得可以将它们归入一个提议命名为环钠菌科(Cyclonatronaceae)的新科。
