Abstract

BACKGROUND

Regulatory T cells (Tregs) play an important role in regulation of immune response and immunologic tolerance in cancer. Gastrointestinal cancer is still a leading cause of cancer-related death in the world. This study aimed to detect Tregs in patients with gastrointestinal cancer.

METHODS

In this study, 45 gastric cancer patients, 50 colorectal cancer patients and 50 healthy controls were enrolled. Flow cytometry was used to detect CD4CD25CD127 Tregs, CD4CD25, and CD4 cells in peripheral blood. Cytokine interleukin-10 (IL-10) and transforming growth factor-β1 (TGF-β1) in peripheral blood and in the supernatant of Tregs cultures were measured by enzyme linked immunosorbent assay.

RESULTS

Compared with healthy controls, the levels of CD4CD25CD127 Tregs and CD4CD25 cells increased significantly in patients with gastrointestinal cancer. Patients with gastrointestinal cancer also showed a significantly increased levels of IL-10 and TGF-β1 in both peripheral blood and CD4CD25CD127 Tregs culture medium.

CONCLUSION

The present study firstly demonstrated that gastrointestinal patients have a compromised immune status where the CD4CD25CD127 Tregs, as well as levels of IL-10 and TGF-β1 are elevated. The data offered new information for understanding the immunological features of gastrointestinal patients, as well as provided new insights into approaches to develop new immunotherapies for patients with gastrointestinal cancer.

背景

调节性 T 细胞(Tregs)在癌症的免疫反应和免疫耐受调节中发挥着重要作用。胃肠道癌仍然是全球癌症相关死亡的主要原因。本研究旨在检测胃肠道癌患者中的 Tregs。

方法

结果

结论

本研究首次证明胃肠道癌患者的免疫状态受损,CD4CD25CD127 Tregs 及其细胞因子 IL-10 和 TGF-β1 水平升高。该数据为了解胃肠道癌患者的免疫学特征提供了新信息,并为开发胃肠道癌患者新的免疫治疗方法提供了新的思路。
