Abstract

OBJECTIVE

An association between microRNAs (miRNAs) and adhesion proteins expression with repeated implantation failure (RIF) has been recently reported; however, these findings are controversial. This study aims to evaluate the endometrial and circulating expressions of miR-145, miR-155-5p, and miR-224 in addition to the endometrial expressions of membrane protein palmitoylated-5 and endothelial cell adhesion molecule-1 in patients with RIF compared to control subjects.

MATERIALS AND METHODS

This case-control study was carried out between June 2021-July 2022. Subjects included 17 patients with RIF and 17 control subjects, who had previous spontaneous term pregnancy with a live birth, who referred to the Medical Centre of Arash Hospital, Tehran, Iran. Endometrial tissue samples were obtained via hysteroscopy and Pipelle catheter in the RIF and control subjects, respectively. Plasma samples were collected after ovulation in all subjects. The expression levels of miR-224, miR-145, and miR-155-5p were evaluated by quantitative real-time polymerase chain reaction (qRT-PCR). The student's t test, chi-square, Mann-Whitney U, and analysis of covariance (ANCOVA) were used for data analyses.

RESULTS

RIF patients had less endometrial miR-155-5p expression, and higher endometrial and circulating expressions of miR-145 and miR-224 compared to control subjects. Endometrial expression significantly decreased in patients with RIF compared to the control group. There was a positive correlation between circulating miR-224 and endometrial miR-155-5p, and between circulating miR-155-5p and endometrial expression levels in patients with RIF.

CONCLUSION

The present study suggests that circulating miR-224, endometrial miR-145, and PECAM-1 can be reliable, novel biomarkers for diagnosis of RIF.

目的

最近有报道称微小RNA(miRNA)与黏附蛋白表达和反复种植失败(RIF)之间存在关联;然而,这些发现存在争议。本研究旨在评估RIF患者与对照受试者相比,miR-145、miR-155-5p和miR-224的子宫内膜及循环表达,以及膜蛋白棕榈酰化-5和内皮细胞黏附分子-1的子宫内膜表达。

材料与方法

本病例对照研究于2021年6月至2022年7月进行。研究对象包括17例RIF患者和17例对照受试者,对照受试者曾有过足月自然分娩活产史,他们前往伊朗德黑兰阿拉什医院医疗中心就诊。分别通过宫腔镜检查和Pipelle导管获取RIF患者和对照受试者的子宫内膜组织样本。所有受试者在排卵后采集血浆样本。通过定量实时聚合酶链反应(qRT-PCR)评估miR-224、miR-145和miR-155-5p的表达水平。采用学生t检验、卡方检验、曼-惠特尼U检验和协方差分析(ANCOVA)进行数据分析。

结果

与对照受试者相比,RIF患者的子宫内膜miR-155-5p表达较低,而miR-145和miR-224的子宫内膜及循环表达较高。与对照组相比,RIF患者的子宫内膜PECAM-1表达显著降低。在RIF患者中,循环miR-224与子宫内膜miR-155-5p之间以及循环miR-155-5p与子宫内膜PECAM-1表达水平之间存在正相关。

结论

本研究表明,循环miR-224、子宫内膜miR-145和PECAM-1可作为诊断RIF的可靠新型生物标志物。
