The purpose of the present study was to study the potential anti-arthritic and antioxidant effects of trehalose in an experimental model of complete Freund's adjuvant (CFA)-induced arthritis.
Arthritis was induced via subcutaneous injection of CFA (0.1) into the right footpad of each rat. Trehalose (10 mg/kg per day) and indomethacin (5 mg/kg) as a reference drug were intraperitoneally injected into CFA-induced arthritic rats from days 0 to 21. Changes in paw volume, pain responses, arthritic score, and oxidative/antioxidative parameters were determined.
Trehalose administration could significantly decrease arthritis scores (p <0.01) and paw edema (p <0.001), and significantly increase the nociceptive threshold (p <0.05) in CFA-induced arthritic rats. Trehalose also significantly reduced the pro-oxidant-antioxidant balance values when compared to CFA treatment alone. In addition, no significant difference was found between the trehalose group and indomethacin as a positive control group.
The current study suggests that trehalose has a protective effect against arthritis, which may be mediated by antioxidative effects of this disaccharide.
本研究旨在研究海藻糖在完全弗氏佐剂(CFA)诱导的关节炎实验模型中的潜在抗关节炎和抗氧化作用。
通过向每只大鼠的右足底皮下注射 CFA(0.1)来诱导关节炎。海藻糖(10mg/kg/天)和吲哚美辛(5mg/kg)作为参考药物,从第 0 天到第 21 天腹腔注射到 CFA 诱导的关节炎大鼠中。测定爪体积、疼痛反应、关节炎评分和氧化/抗氧化参数的变化。
海藻糖给药可显著降低关节炎评分(p<0.01)和爪肿胀(p<0.001),并显著提高 CFA 诱导的关节炎大鼠的痛觉阈值(p<0.05)。与单独 CFA 处理相比,海藻糖还显著降低了促氧化剂-抗氧化剂平衡值。此外,海藻糖组与阳性对照药物吲哚美辛之间未发现显著差异。
本研究表明,海藻糖对关节炎具有保护作用,这可能是通过这种二糖的抗氧化作用介导的。
