BACKGROUND
Tuberculosis (TB) is one of the leading causes of death worldwide. Besides, one-third of the world population is infected with (MTB) while staying clinically asymptomatic; the situation is called latent TB infection (LTBI). MiR-21, miR-31, miR-146a, and miR-155 play an important role in many immune and inflammatory pathways. In the present study the expression levels of MiR-21, miR-31, miR-146a, and miR-155 in peripheral blood mononuclear cells (PBMCs) from patients with active TB, latently infected individuals (LTBI), and healthy controls (HC) were investigated. Participants were recruited at the Bouali Hospital, Zahedan University of Medical Sciences, Zahedan, Iran from 2010 to 2011.
METHODS
PBMCs were stimulated with PPD before RNA extraction. TaqMan RT-qPCR assay was used to analyze the expression levels of miRNAs.
RESULTS
The results indicated no significant differences in the expression of miR-21 and miR-31 between different groups; however, in patients with active TB, the expression of miR-21 (=0.03) and miR-31 (=0.04) were significantly increased after stimulation with PPD compared to the unstimulated condition. The expression of miR-146 in response to PPD in both LTBI (=0.02) and TB (=0.03) groups compared to the HC group was increased. No significant differences were found in the expression level of miR-155 in response to PPD between LTBI and HC groups. However, the fold change was significantly higher in the TB group in comparison with the HC (=0.03) and LTBI (=0.05) groups.
CONCLUSION
The results confirm the main role of miR-146 and miR-155 in TB infection and suggest a role for miR-146 and miR-155 as infection and activation markers in tuberculosis infection, respectively.

结核病(TB)是全球主要死因之一。此外,全球三分之一的人口感染了结核分枝杆菌(MTB),但临床上并无症状,这种情况被称为潜伏性结核感染(LTBI)。MiR - 21、MiR - 31、MiR - 146a和MiR - 155在许多免疫和炎症途径中发挥重要作用。在本研究中,对活动性结核病患者、潜伏感染个体(LTBI)和健康对照(HC)外周血单个核细胞(PBMCs)中MiR - 21、MiR - 31、MiR - 146a和MiR - 155的表达水平进行了研究。参与者于2010年至2011年在伊朗扎赫丹医科大学布阿利医院招募。
方法
在提取RNA之前,用结核菌素纯蛋白衍生物(PPD)刺激PBMCs。采用TaqMan RT - qPCR测定法分析微小RNA(miRNAs)的表达水平。
结果
结果表明,不同组之间MiR - 21和MiR - 31的表达无显著差异;然而,在活动性结核病患者中,与未刺激状态相比,用PPD刺激后MiR - 21(=0.03)和MiR - 31(=0.04)的表达显著增加。与健康对照组相比,LTBI组(=0.02)和结核病组(=0.03)中MiR - 146对PPD刺激的反应表达均增加。LTBI组和健康对照组之间,MiR - 155对PPD刺激的反应表达水平无显著差异。然而,与健康对照组(=0.03)和LTBI组(=0.05)相比,结核病组的倍数变化显著更高。
结论
结果证实了MiR - 146和MiR - 155在结核病感染中的主要作用,并分别提示MiR - 146和MiR - 155作为结核病感染中的感染和激活标志物的作用。
