We studied the effect of selective α-adrenergic receptor antagonist JP-1302 in concentrations of 10-10 M on inotropy, chronotropy, and coronary flow in the Langendorff-isolated rat heart. JP-1302 in all studied concentrations decreased the left-ventricular myocardium force contraction, HR, and coronary flow. The maximum inotropic, chronotropic, and vascular effects were observed when the antagonist was applied to the perfused solution in a concentration of 10 M. The least pronounced decrease in the studied parameters was observed at JP-1302 concentrations of 10 and 10 M. The obtained data indicate the participation of this subtype of α-adrenergic receptors in the regulation of activity of isolated adult rats heart.
我们研究了浓度为10⁻¹⁰ M的选择性α-肾上腺素能受体拮抗剂JP-1302对Langendorff离体大鼠心脏的心肌收缩力、心率和冠状动脉血流量的影响。在所研究的所有浓度下,JP-1302均降低了左心室心肌收缩力、心率和冠状动脉血流量。当拮抗剂以10⁻⁸ M的浓度加入到灌注液中时,观察到最大的心肌收缩力、心率和血管效应。在JP-1302浓度为10⁻¹⁰ M和10⁻⁹ M时,所研究参数的下降最为不明显。所得数据表明该亚型α-肾上腺素能受体参与了成年离体大鼠心脏活动的调节。
