Abstract

A Gram-staining negative, non-motile, rod-shaped, oxidase negative and catalase positive strain WL0021 was isolated from cricket (Gryllus chinensis) living in the campus of Hohai University. Strain WL0021 was characterized utilizing a polyphasic taxonomy approach. The major fatty acids (> 5%) for strain WL0021 were C and summed feature 8, and the major polar lipids were diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, phosphatidylcholine, phospholipid, two aminolipids, and an unidentified polar lipid. Ubiquinone-10 was detected as the predominant respiratory quinone. The results of 16S rRNA gene phylogenetic analyses revealed that strain WL0021 had the highest sequence similarity of 95.3% to Microvirga flavescens c27j1 and strain WL0021 formed a distinct linage within the family Methylobacteriaceae in the phylogenetic trees. Whole genomic DNA G+C content was 48.3%. Combined with the results from this study, strain WL0021 should represent a novel genus in the family Methylobacteriaceae, for which the name Hohaiivirga grylli gen. nov., sp. nov. (type strain WL0021=GDMCC 1.2420 =JCM 34655=MCCC 1K05886) is proposed.

从生活在河海大学校园里的蟋蟀(Gryllus chinensis)中分离到一株革兰氏染色阴性、非运动、杆状、氧化酶阴性和过氧化氢酶阳性的菌株 WL0021。利用多相分类学方法对菌株 WL0021 进行了表征。菌株 WL0021 的主要脂肪酸(>5%)为 C 和特征脂肪酸 8,主要极性脂为双磷脂酰甘油、磷脂酰乙醇胺、磷脂酰甘油、磷脂酰胆碱、磷脂、两种氨基脂和一种未鉴定的极性脂。检测到泛醌-10 作为主要的呼吸醌。16S rRNA 基因系统发育分析的结果表明,菌株 WL0021 与 Microvirga flavescens c27j1 的序列相似性最高,为 95.3%,菌株 WL0021 在系统发育树中与 Methylobacteriaceae 家族中的其他菌株形成了一个独特的分支。全基因组 DNA G+C 含量为 48.3%。结合本研究结果,菌株 WL0021 应代表 Methylobacteriaceae 家族中的一个新属,因此建议将其命名为 Hohaiivirga grylli gen. nov., sp. nov.(模式菌株 WL0021=GDMCC 1.2420=JCM 34655=MCCC 1K05886)。
