A thermophilic cellulase-producing bacterium, strain HSW-8, isolated from hot spring waters in South Korea, was subjected to a taxonomic analysis. Cells of strain HSW-8 were gram-stain-negative, facultatively anaerobic, rod-shaped, with optimum growth at 45 °C, pH 7.0, in the presence of 0% (w/v) NaCl. Strain HSW-8 showed the highest 16S rRNA gene sequence similarity to Sinimarinibacterium flocculans NH6-24 (97.52%), followed by Fontimonas thermophila DSM 23609 (96.97%), Solimonas flava CW-KD 4 (95.24%), and Solimonas variicoloris DSM 15731 (95.18%). Based on 16S rRNA phylogeny, strain HSW-8 is phylogenetically closely related to Fontimonas thermophila DSM 23609 and Sinimarinibacterium flocculans DSM 104150 and could be distinguished from the type species based on their phenotypic properties. The genome length of strain HSW-8 was 3.32 Mbp with a 67.33% G + C content. The average nucleotide identity and digital DNA-DNA hybridization values between strain HSW-8 and its closely related type strains were 75.4-83.2 and 20.2-26.2%, respectively. Summed feature 8 (Cω7c and/or Cω6c), C and iso-C identified the major fatty acids (> 10%). Phosphatidylglycerol and phosphatidylethanolamine were demonstrated as the major polar lipids while the respiratory quinone is ubiquinone-8. Strain HSW-8 exhibited multiple adaptations for survival at high temperatures, including diverse potential motility mechanisms and toxin-antitoxin systems, as evidenced by both phenotypic characteristics and genomic analysis. Based on genotypic and phenotypic features, strain HSW-8 (= KCTC 92765 = GDMCC 1.4313) represents a novel Sinimarinibacterium species, in which the name Sinimarinibacterium thermocellulolyticum sp. nov. is proposed.
从韩国温泉水中分离出的一株嗜热产纤维素酶细菌HSW-8,进行了分类学分析。HSW-8菌株细胞革兰氏染色阴性,兼性厌氧,杆状,在45°C、pH 7.0、0%(w/v)NaCl条件下生长最佳。HSW-8菌株与絮凝中华海菌NH6-24的16S rRNA基因序列相似性最高(97.52%),其次是嗜热丰蒂莫氏菌DSM 23609(96.97%)、黄色索利莫氏菌CW-KD 4(95.24%)和变色索利莫氏菌DSM 15731(95.18%)。基于16S rRNA系统发育,HSW-8菌株在系统发育上与嗜热丰蒂莫氏菌DSM 23609和絮凝中华海菌DSM 104150密切相关,可根据其表型特征与模式种区分开来。HSW-8菌株的基因组长度为3.32 Mbp,G+C含量为67.33%。HSW-8菌株与其密切相关的模式菌株之间的平均核苷酸同一性和数字DNA-DNA杂交值分别为75.4-83.2%和20.2-26.2%。总和特征8(Cω7c和/或Cω6c)、C和异-C鉴定出主要脂肪酸(>10%)。磷脂酰甘油和磷脂酰乙醇胺被证明是主要极性脂质,而呼吸醌为泛醌-8。HSW-8菌株表现出多种在高温下生存的适应性,包括多种潜在的运动机制和毒素-抗毒素系统,这在表型特征和基因组分析中均有体现。基于基因型和表型特征,HSW-8菌株(=KCTC 92765=GDMCC 1.4313)代表一个新的中华海菌物种,其中提出了热解纤维素中华海菌新种的名称。
