Abstract

OBJECTIVE

The study aimed to investigate mutations and mRNA expression of the gene in resistant isolates.

METHODS

This cross-sectional study was conducted from October 2018 to June 2019. High vaginal swab samples were collected from Hayatabad Medical Complex and transported to Khyber Medical University. Samples were inoculated on different media and identified by 20C AUX strips. Antifungal susceptibility was determined using the disc diffusion and broth microdilution methods. The was amplified and sequenced to find amino acid polymorphisms. Real-time PCR was performed to study level of expression.

RESULTS

A total of seventy-three (n=73) C out of 369 samples were isolated. Among the isolates 49.3%, 54.8%, 53.4%, 47.9%, 30.1% were resistant to fluconazole, Clotrimazole, Miconazole, Voriconazole and Itraconazole, respectively. Sanger sequencing of gene of isolates revealed six synonymous mutations. Expression level of mRNA of gene in azoles sensitive stains (3.72±2.22) was higher than those in the resistant strains (1.74±0.96).

CONCLUSION

This study revealed synonymous mutations and low expression of gene in azole-resistant .

目的

本研究旨在调查耐药菌株中该基因的突变情况及mRNA表达。

方法

本横断面研究于2018年10月至2019年6月进行。从哈亚塔巴德医疗中心采集高阴道拭子样本,并运送至开伯尔医科大学。样本接种于不同培养基上,并用20C AUX试纸条进行鉴定。采用纸片扩散法和肉汤微量稀释法测定抗真菌药敏性。对该基因进行扩增和测序以发现氨基酸多态性。进行实时PCR以研究该基因的表达水平。

结果

369份样本中共分离出73株念珠菌。在分离菌株中,分别有49.3%、54.8%、53.4%、47.9%、30.1%对氟康唑、克霉唑、咪康唑、伏立康唑和伊曲康唑耐药。对分离菌株的该基因进行桑格测序,发现6个同义突变。该基因在唑类敏感菌株中的mRNA表达水平(3.72±2.22)高于耐药菌株(1.74±0.96)。

结论

本研究揭示了唑类耐药念珠菌中该基因的同义突变及低表达。
