Cumin seeds were fractionated with various solvents to evaluate their antioxidant activity and anti-adipogenic effects in 3T3-L1 adipocytes. In vitro antioxidant assays such as DPPH, ABTS cation scavenger, FRAP, ORAC, and TPC, and 3T3-L1 adipocytes differentiation were performed. The results of these assays show that ethyl acetate fraction of cumin seeds (CSEA) had significantly superior antioxidant activity compared to other cumin seeds fractions. Thymol, benzyl alcohol, and 2-methyl-3-phenylpropanal were analyzed as major volatile components in CSEA. Treatment of 50 and 75 μg/mL CSEA inhibited fat accumulation in 3T3-L1 adipocytes significantly ( < 0.05). When 3T3-L1 adipocytes were treated with CSEA at 75 μg/mL, the expressions of genes and proteins related to fat differentiation and accumulation were significantly reduced ( < 0.05). Therefore, CSEA could be used as antioxidants and as a functional ingredient in anti-adipogenic formulations.
用各种溶剂对孜然种子进行分级分离,以评估其在3T3-L1脂肪细胞中的抗氧化活性和抗脂肪生成作用。进行了体外抗氧化试验,如DPPH、ABTS阳离子清除剂、FRAP、ORAC和TPC试验,以及3T3-L1脂肪细胞分化试验。这些试验结果表明,孜然种子的乙酸乙酯级分(CSEA)与其他孜然种子级分相比具有显著优越的抗氧化活性。分析得出百里香酚、苯甲醇和2-甲基-3-苯基丙醛是CSEA中的主要挥发性成分。用50和75μg/mL的CSEA处理可显著抑制3T3-L1脂肪细胞中的脂肪积累(P<0.05)。当用75μg/mL的CSEA处理3T3-L1脂肪细胞时,与脂肪分化和积累相关的基因和蛋白质表达显著降低(P<0.05)。因此,CSEA可作为抗氧化剂和抗脂肪生成制剂中的功能性成分。
