Abstract

A flexible silver triangular nanoplate (Ag TNPs)@m-phenylenediamine (mPD)@polyacrylonitrile (PAN) skin patch based on electrospinning is fabricated and applied to detect human beta-defensin 2 (HBD-2) by using surface-enhanced Raman scattering (SERS). First, PAN was used as the base material to create a fibrous membrane through electrospinning. The fibers were then modified with Ag TNPs by mPD endowing positive charges, forming a flexible Ag TNPs@mPD@PAN substrate. The flexible substrate features advantages of simple preparation, low cost, and convenient portability, which are suitable for in situ rapid detection on irregular surfaces. The combination of the flexible Ag TNPs@mPD@PAN substrate with Ag TNPs probes enabled the specific detection of HBD-2. It demonstrated satisfactory sensitivity and reproducibility with a wide detection range of 100-10 ng/mL and a limit of detection (LOD) of 59.57 fg/mL. Eventually, the flexible substrate was applied to HBD-2 detection on the surface of contaminated chicken skin. This study provides a simple and highly sensitive method for the detection of HBD-2, offering a novel approach for the non-invasive rapid diagnosis of psoriasis and the monitoring of disease progression.

基于静电纺丝技术制备了一种柔性银三角纳米片(Ag TNPs)@间苯二胺(mPD)@聚丙烯腈(PAN)皮肤贴片,并利用表面增强拉曼散射(SERS)技术用于检测人β-防御素2(HBD-2)。首先,以PAN为基材,通过静电纺丝制备纤维膜。然后,利用带正电荷的mPD对纤维进行Ag TNPs修饰,形成柔性Ag TNPs@mPD@PAN基底。该柔性基底具有制备简单、成本低、便于携带等优点,适用于在不规则表面进行原位快速检测。柔性Ag TNPs@mPD@PAN基底与Ag TNPs探针相结合,实现了对HBD-2的特异性检测。其在100 - 10 ng/mL的宽检测范围内表现出令人满意的灵敏度和重现性,检测限(LOD)为59.57 fg/mL。最终,将该柔性基底应用于污染鸡皮表面的HBD-2检测。本研究为HBD-2的检测提供了一种简单且高灵敏度的方法,为银屑病的无创快速诊断和疾病进展监测提供了一种新途径。
