Abstract

Reactivation of single-stranded DNA phage, photodynamically inactivated in the presence of proflavine sulfate, by three isogenic Escherichia coli strains having different DNA repair capabilities has been studied. It was found that reactivation of photoinactivated phiX174 was possible only if the host cells were recombination proficient (recA(+)) and had been lightly irradiated with UV light prior to infection; the presence of the uvrA(+) gene was not essential. Only a small part of the proflavine-mediated photodynamic damage in phiX174 could be repaired in this fashion. Burst sizes of reactivated phages were, however, comparable to those of normal unirradiated phages.

研究了三种具有不同DNA修复能力的同基因大肠杆菌菌株对在硫酸原黄素存在下经光动力灭活的单链DNA噬菌体的复活作用。结果发现,只有宿主细胞具有重组能力(recA(+))且在感染前经紫外线轻度照射,光灭活的φX174噬菌体才有可能复活;uvrA(+)基因的存在并非必需。通过这种方式,只能修复φX174中一小部分由原黄素介导的光动力损伤。然而,复活噬菌体的爆发量与正常未照射的噬菌体相当。
