Native myosin was obtained from skeletal and smooth muscles and purified as much as possible. Rabbits were immunized with myosin preparations. Antiserum specificity was tested by double diffusion in agarose and by the indirect immunofluorescence technique. Application of the latter technique enabled detecting ballast antibodies in the immune sera. Monospecific antisera with high enough titer were obtained after cross-immunosorption by sorbents from 0.6 M extracts of skeletal and smooth muscles.
