DNA of leprosy cells in serial paraffine sections stained after Felgen were studied photometrically in scanning microscope SPM with electron computer Wang 720C. Significant differences in extinction values (up to 30--35%) between individual sections within the same series were revealed. Cumulative results of different series were rather uniform. It was concluded that photometric studies of serial sections should be carried out for reliable determination of DNA.
对经费尔根染色后的连续石蜡切片中的麻风细胞DNA,使用王720C电子计算机在扫描显微镜SPM上进行了光度测定研究。结果显示,同一系列内各个切片之间的消光值存在显著差异(高达30%-35%)。不同系列的累积结果相当一致。得出的结论是,为可靠地测定DNA,应对连续切片进行光度研究。
