To determine the mRNA expression levels of copper-zinc superoxide dismutase (Cu, Zn-SOD) and manganese SOD (Mn-SOD) in healthy and inflamed human dental pulp tissue.
Sixteen patients with symptomatic irreversible pulpitis (eight females and eight males) were selected for study. Normal healthy pulps were removed from extracted mandibular third molar teeth from 10 systemically healthy individuals (six females and four males). QRT-PCR analysis of Cu, Zn-SOD and Mn-SOD mRNA expression was carried out in 16 cases of irreversible pulpitis and in 10 cases of systemically healthy donors. The Shapiro-Wilk's test was used to test the normality of data, whereas the Mann-Whitney U-test was used to evaluate the significance of the differences between groups. Differences in the expression levels were considered to be statistically significant for P-values <0.05.
A significant increase (P < 0.05) occurred in both Cu, Zn-SOD and Mn-SOD mRNA expression in cases of irreversible pulpitis. The increase in Mn-SOD level was significantly higher (P < 0.05) than the change observed for Cu, Zn-SOD.
The development of pulpitis is associated with elevated transcription of both Cu, Zn-SOD and Mn-SOD; pulp tissue inflammation generated higher Mn-SOD transcription compared with Cu, Zn-SOD.
测定健康和炎症状态下人类牙髓组织中铜锌超氧化物歧化酶(Cu,Zn-SOD)和锰超氧化物歧化酶(Mn-SOD)的mRNA表达水平。
选取16例有症状的不可逆性牙髓炎患者(8例女性和8例男性)进行研究。从10名全身健康个体(6例女性和4例男性)拔除的下颌第三磨牙中获取正常健康牙髓。对16例不可逆性牙髓炎患者和10例全身健康供体进行Cu,Zn-SOD和Mn-SOD mRNA表达的定量逆转录聚合酶链反应(QRT-PCR)分析。采用夏皮罗-威尔克检验(Shapiro-Wilk's test)检测数据的正态性,而采用曼-惠特尼U检验(Mann-Whitney U-test)评估组间差异的显著性。当P值<0.05时,表达水平的差异被认为具有统计学意义。
不可逆性牙髓炎患者中Cu,Zn-SOD和Mn-SOD mRNA表达均显著增加(P<0.05)。Mn-SOD水平的增加显著高于Cu,Zn-SOD的变化(P<0.05)。
牙髓炎的发生与Cu,Zn-SOD和Mn-SOD的转录升高有关;牙髓组织炎症产生的Mn-SOD转录高于Cu,Zn-SOD。
