The aim of this study was to investigate whether local delivery of alendronate could improve bone formation after bioactive glass grafting in rat mandible.
Twenty-six male Sprague-Dawley rats were divided into control and experimental groups (13 rats/group). A surgical defect was created on the angle of mandible of each animal. In the experimental group, a bioactive glass soaked with the alendronate solution was placed in the bone defect, and in the control group the bioactive glass soaked with saline was used. All animals were killed after 4 weeks. The number of osteoclasts and the amount of new bone formation were evaluated and compared.
Four weeks after surgery, the experimental group had significantly more bone formation than the control groups (P < .05). However, no statistically significant difference was found between the groups when the numbers of osteoclasts were compared.
Histologic results showed that a single dose of local delivery of alendronate improves bone formation. However, further studies are required to elucidate the effect of local delivery of alendronate on bone formation in humans.
本研究旨在探讨阿仑膦酸钠局部给药是否能改善大鼠下颌骨生物活性玻璃移植后的骨形成。
将26只雄性Sprague-Dawley大鼠分为对照组和实验组(每组13只大鼠)。在每只动物的下颌角制造一个手术缺损。实验组将浸泡过阿仑膦酸钠溶液的生物活性玻璃置于骨缺损处,对照组则使用浸泡过生理盐水的生物活性玻璃。4周后处死所有动物。评估并比较破骨细胞数量和新骨形成量。
术后4周,实验组的骨形成明显多于对照组(P <.05)。然而,比较两组破骨细胞数量时未发现统计学上的显著差异。
组织学结果表明,单次局部给予阿仑膦酸钠可改善骨形成。然而,需要进一步研究以阐明阿仑膦酸钠局部给药对人类骨形成的影响。
