To explore the mechanism of Xiaotan Sanjie Recipe (XtSjR, Recipe for dissolving phlegm to eliminate stagnation) in inhibiting proliferation of gastric cancer cells.
The nude mouse human gastric cancer MKN-45 in situ transplantation tumor model was established by use of OB glue, and 40 model mice were randomized into 5 groups, model group, low-dose XtSjR group, middle-dose XtSjR group, high-dose XtSjR group, and 5-Fu group, 8 rats in each group. Human gastric cancer MKN-45 telomerase reverse transcriptase (hTERT) protein and mRNA expressions were assayed by immunohistochemical method and real-time fluorescence quantitative RT-PCR, and influences of XtSjR on the expressions of hTERT protein and mRNA were investigated in the nude mouse human gastric cancer MKN-45 in situ tumor transplantation model.
探讨消痰散结方抑制胃癌细胞增殖的作用机制。
采用 OB 胶建立裸鼠人胃癌 MKN-45 原位移植瘤模型,将 40 只模型鼠随机分为 5 组,即模型组、消痰散结方低、中、高剂量组和 5-Fu 组,每组 8 只。采用免疫组化法和实时荧光定量 RT-PCR 法检测人胃癌 MKN-45 端粒酶逆转录酶(hTERT)蛋白和 mRNA 的表达,观察消痰散结方对裸鼠人胃癌 MKN-45 原位移植瘤模型中 hTERT 蛋白和 mRNA 表达的影响。
1)消痰散结方低、中、高剂量组与模型组比较,平均瘤重均有显著差异(均 P < 0.01);2)消痰散结方中、高剂量组 hTERT 阳性表达率与模型组比较,均有显著差异(P < 0.05 或 P < 0.01);3)消痰散结方中、高剂量组 hTERT mRNA 含量与模型组比较,均有显著差异(P < 0.05 或 P < 0.01)。
1)消痰散结方能显著抑制胃癌细胞的生长;2)消痰散结方通过下调 hTERT 蛋白和 mRNA 的表达,抑制端粒酶活性,逐渐缩短癌细胞端粒的长度,使其丧失无限增殖的能力,从而最终抑制肿瘤细胞的生长和增殖。
