Abstract

BACKGROUND & AIM: Dyslipidemia is one of the major complications in patients with type 2 diabetes mellitus (T2DM). Dietary fat intake and genetic factors including CETP Taq1B polymorphism could also affect lipid profile concentrations, in particular HDL-c. We decided to study the frequency of this polymorphism and its interaction with dietary fat intake on HDL-c concentration among Iranian T2DM patients with and without dyslipidemia.

METHODS

In this comparative study, serum samples were collected from 55 patients with dyslipidemia and 129 patients without dyslipidemia. Validated semi-quantitative FFQ was used for food consumption data. CETP Taq1B polymorphism was studied by polymerase chain reaction-restriction length polymorphism (PCR-RFLP). We used χ and two-way ANOVA tests for statistical analysis.

RESULTS

The frequency of B1B1 genotype was higher in patients with dyslipidemia (p = 0.01). There was no significant relationship between CETP Taq1B polymorphism and lipid profile concentrations. In patients without dyslipidemia, the interaction between the polymorphism and total fat intake on HDL-c concentration as well as TG/HDL ratio was significant (p = 0.02 and p = 0.009 respectively). This was more evident in B1B1 genotype. Moreover, HDL-c concentration was significantly higher in B2B2 genotype with low total fat intake.

CONCLUSION

Higher total fat intake may affect the relationship between CETP Taq1B polymorphism and HDL-c concentration in patients with normolipidemic T2DM.

血脂异常是 2 型糖尿病(T2DM)患者的主要并发症之一。膳食脂肪摄入和包括 CETP Taq1B 多态性在内的遗传因素也可能影响血脂谱浓度,尤其是 HDL-c。我们决定研究该多态性的频率及其与伊朗 T2DM 患者血脂异常和无血脂异常患者的饮食脂肪摄入对 HDL-c 浓度的相互作用。

方法

在这项比较研究中,从 55 名血脂异常患者和 129 名无血脂异常患者中采集血清样本。使用经过验证的半定量 FFQ 收集食物消费数据。通过聚合酶链反应-限制性长度多态性(PCR-RFLP)研究 CETP Taq1B 多态性。我们使用 χ 和双向 ANOVA 检验进行统计分析。

结果

血脂异常患者 B1B1 基因型的频率更高(p=0.01)。CETP Taq1B 多态性与血脂谱浓度之间没有显著关系。在无血脂异常的患者中,多态性与总脂肪摄入量对 HDL-c 浓度以及 TG/HDL 比值的相互作用具有显著性(分别为 p=0.02 和 p=0.009)。在 B1B1 基因型中更为明显。此外,B2B2 基因型的总脂肪摄入量低时,HDL-c 浓度显著升高。

结论

总脂肪摄入量较高可能会影响正常血脂 T2DM 患者 CETP Taq1B 多态性与 HDL-c 浓度之间的关系。
