Abstract

AIM

To evaluate the effects of aqueous extract of Salep on Paraquat-mediated liver injury.

METHODS

In this experimental study, 56 adult male Wistar rats were divided randomly to 7 groups as control, sham, and 5 experimental groups. In control group, rats did not receive any substance during experiment. In Sham group, rats were given distilled water according to their body weight and in experimental groups, Paraquat alone and with different doses of Salep aqueous extract (40, 80, 160 and 320 mg/kg) was given intraperitoneal daily for 14 d. After that, liver biochemical parameter and histologic changes were analyzed and compared in different groups.

RESULTS

Paraquat compared to control and sham groups, significantly ( < 0.05) increased serum level of alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), bilirubin, malondialdehyde (MDA) and total oxidant capacity (TOC); while level of total protein, albumin and total antioxidant capacity (TAC) were remarkably decreased by Paraquat. Salep at doses of 80, 160 and 320 mg/kg significantly decreased serum level of ALT, AST, ALP, bilirubin, MDA and TOC and significantly increased total protein, albumin and TAC level as compared to Paraquat exposed group in dose dependent manner. Aqueous extract of Salep at doses of 40 mg/kg made no significant changes in serum level of mentioned biochemical parameters. Liver microscopic observation revealed that Paraquat could cause hepatocyte necrosis, degenerative changes, proliferation and activation of Kupffer cells (sporadically) which were reduced by Salep treatment.

CONCLUSION

Salep possesses remarkable hepatoprotection activity against Paraquat-induced hepatic injury by having antioxidant activity and reducing lipid peroxidation and oxidative stress.

目的

评估鼠尾草提取物对百草枯介导的肝损伤的影响。

方法

在本实验研究中,将56只成年雄性Wistar大鼠随机分为7组,即对照组、假手术组和5个实验组。对照组大鼠在实验期间不接受任何物质。假手术组大鼠按体重给予蒸馏水,实验组大鼠每天腹腔注射单独的百草枯以及不同剂量的鼠尾草提取物(40、80、160和320毫克/千克),持续14天。之后,分析并比较不同组的肝脏生化参数和组织学变化。

结果

与对照组和假手术组相比,百草枯显著(<0.05)升高了血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、碱性磷酸酶(ALP)、胆红素、丙二醛(MDA)和总氧化能力(TOC)的水平;而百草枯显著降低了总蛋白、白蛋白和总抗氧化能力(TAC)的水平。与百草枯暴露组相比,80、160和320毫克/千克剂量的鼠尾草提取物以剂量依赖的方式显著降低了血清ALT、AST、ALP、胆红素、MDA和TOC的水平,并显著提高了总蛋白、白蛋白和TAC的水平。40毫克/千克剂量的鼠尾草提取物对上述生化参数的血清水平没有显著影响。肝脏显微镜观察显示,百草枯可导致肝细胞坏死、退行性变化、库普弗细胞的增殖和激活(散在性),而鼠尾草提取物治疗可减轻这些变化。

结论

鼠尾草通过具有抗氧化活性、减少脂质过氧化和氧化应激,对百草枯诱导的肝损伤具有显著的肝保护活性。
