A Gram-stain-negative, non-motile, non-spore-forming, rod-shaped, aerobic bacterial strain, designated 16F7C-2, was isolated from the Han River, South Korea, and was characterized taxonomically using a polyphasic approach. Comparative 16S rRNA gene sequence analysis showed that strain 16F7C-2 belonged to the family Cytophagaceae in the phylum Bacteroidetes and was most closely related to Hymenobacter algoricola VUG-A23a (98.3%) and Hymenobacter fastidiosus VUG-A124 (97.7%). The G + C content of the genomic DNA of strain 16F7C-2 was 63.4 mol%. The detection of menaquinone MK-7 as the predominant respiratory quinone; a fatty acid profile with summed feature 3 (C ω7c/C ω6c; 19.5%), summed feature 4 (C iso I/C anteiso B; 15.9%), C iso (12.6%), C ω5c (10.5%), and C anteiso (10.4%) as the major components; and a polar lipid profile with phosphatidylethanolamine as the major component also supported the affiliation of strain 16F7C-2 to the genus Hymenobacter. The DNA-DNA relatedness between strain 16F7C-2 and H. algoricola JCM 17214 and H. fastidiosus JCM17224 were 45.2 ± 5.8 and 40.3 ± 2.9%, respectively, clearly showing that the isolate constitutes a new genospecies. Strain 16F7C-2 could be clearly differentiated from its closest neighbors on the basis of its phenotypic, genotypic, and chemotaxonomic features. Therefore, strain 16F7C-2 represents a novel species of the genus Hymenobacter, for which the name Hymenobacter knuensis sp. nov. is proposed. The type strain is 16F7C-2 (=KCTC 52538 = JCM 31814).
从韩国汉江分离出一株革兰氏阴性、无运动性、不形成芽孢、杆状、需氧的细菌菌株,命名为16F7C - 2,并采用多相分类法对其进行分类学特征分析。16S rRNA基因序列比较分析表明,菌株16F7C - 2属于拟杆菌门噬细胞菌科,与嗜冷膜杆菌VUG - A23a(98.3%)和苛求膜杆菌VUG - A124(97.7%)亲缘关系最近。菌株16F7C - 2基因组DNA的G + C含量为63.4 mol%。检测到主要呼吸醌为甲基萘醌MK - 7;脂肪酸谱以特征性总和3(C ω7c/C ω6c;19.5%)、特征性总和4(C异戊酸I/C反异戊酸B;15.9%)、C异戊酸(12.6%)、C ω5c(10.5%)和C反异戊酸(10.4%)为主要成分;极性脂质谱以磷脂酰乙醇胺为主要成分,这些也支持菌株16F7C - 2属于膜杆菌属。菌株16F7C - 2与嗜冷膜杆菌JCM 17214和苛求膜杆菌JCM17224之间的DNA - DNA相关性分别为45.2 ± 5.8%和40.3 ± 2.9%,清楚地表明该分离株构成一个新的基因种。菌株16F7C - 2可根据其表型、基因型和化学分类特征与最接近亲缘种明显区分开来。因此,菌株16F7C - 2代表膜杆菌属的一个新物种,提议将其命名为庆熙膜杆菌(Hymenobacter knuensis sp. nov.)。模式菌株为16F7C - 2(=KCTC 52538 = JCM 31814)。
