A Gram-stain negative, aerobic, short rod-shaped, motile by flagella bacterial strain (THG-N2.35), was isolated from Pu'er tea. Growth occurred at 10-40 °C (optimum 28 °C), at pH 4-7 (optimum 7) and at 0-5% NaCl (optimum 1%). Based on 16S rRNA gene sequence analysis, the near phylogenetic neighbours of strain THG-N2.35 were identified as Paracoccus hibisci KACC 18632 (99.0%), Paracoccus tibetensis CGMCC 1.8925 (98.7%), Paracoccus beibuensis CGMCC 1.7295 (98.2%), Paracoccus aestuarii KCTC 22049 (98.2%), Paracoccus rhizosphaerae LMG 26205 (98.1%), Paracoccus zeaxanthinifaciens ATCC 21588 (97.1%), Paracoccus marcusii DSM 11574 (97.0%). Levels of similarity between strain THG-N2.35 and other Paracoccus species were lower than 97.0%. DNA-DNA hybridization values between strain THG-N2.35 and P. hibisci KACC 18632, P. tibetensis CGMCC 1.8925, P. beibuensis CGMCC 1.7295, P. aestuarii KCTC 22049, P. rhizosphaerae LMG 26205, P. zeaxanthinifaciens ATCC 21588, P.marcusii DSM 11574 were 47.5% (42.3%, reciprocal analysis), 36.1% (32.3%), 24.7% (22.1%), 19.2% (16.3%), 11.3% (8.8%), 11.1% (10.8%), 6.1% (5.8%), respectively. The DNA G+C content of strain THG-N2.35 was 62.3 mol%. The polar lipids were diphosphatidylglycerol, phosphatidyl-N-methylethanolamine, phosphatidylethanolamine, phosphatidylglycerol and phosphatidylcholine. The quinone was ubiquinone-10 (Q-10). The major fatty acids were C 3OH, C, C and C ω7ϲ. On the basis of the phylogenetic analysis, chemotaxonomic data, physiological characteristics and DNA-DNA hybridization data, strain THG-N2.35 represent a novel species of the genus Paracoccus, for which the name Paracoccus pueri sp. nov. is proposed. The type strain is THG-N2.35 (= KACC 18934 = CCTCC AB 2016177).
从普洱茶中分离出一株革兰氏染色阴性、需氧、短杆状、具鞭毛能运动的细菌菌株(THG-N2.35)。该菌株在10-40℃(最适温度28℃)、pH 4-7(最适pH 7)和0-5% NaCl(最适浓度1%)条件下生长。基于16S rRNA基因序列分析,菌株THG-N2.35的近缘系统发育邻种被鉴定为 Hibisci 副球菌KACC 18632(相似度99.0%)、西藏副球菌CGMCC 1.8925(98.7%)、北部湾副球菌CGMCC 1.7295(98.2%)、河口副球菌KCTC 22049(98.2%)、根际副球菌LMG 26205(98.1%)、产玉米黄质副球菌ATCC 21588(97.1%)、马库斯副球菌DSM 11574(97.0%)。菌株THG-N2.35与其他副球菌属物种的相似度低于97.0%。菌株THG-N2.35与 Hibisci 副球菌KACC 18632、西藏副球菌CGMCC 1.8925、北部湾副球菌CGMCC 1.7295、河口副球菌KCTC 22049、根际副球菌LMG 26205、产玉米黄质副球菌ATCC 21588、马库斯副球菌DSM 11574的DNA-DNA杂交值分别为47.5%(反向分析为42.3%)、36.1%(32.3%)、24.7%(22.1%)、19.2%(16.3%)、11.3%(8.8%)、11.1%(10.8%)、6.1%(5.8%)。菌株THG-N2.35的DNA G+C含量为62.3 mol%。极性脂为双磷脂酰甘油、磷脂酰-N-甲基乙醇胺、磷脂酰乙醇胺、磷脂酰甘油和磷脂酰胆碱。醌为泛醌-10(Q-10)。主要脂肪酸为C 3OH、C、C和C ω7ϲ。基于系统发育分析、化学分类数据、生理特征和DNA-DNA杂交数据,菌株THG-N2.35代表副球菌属的一个新物种,提议将其命名为普洱茶副球菌(Paracoccus pueri sp. nov.)。模式菌株为THG-N2.35(=KACC 18934 = CCTCC AB 2016177)。
