The objective of this animal study was to promote East Java propolis as a potential natural intracanal medicament for periapical chronic apical periodontitis bone resorption through evaluating the expression of osteoprotegrin (OPG) and osteoclast level.
Propolis extract was produced using a maceration procedure. Thirty Wistar rats were divided into three groups. In group I, the control group, the first upper right molar constituted a healthy tooth. In group II, containing rodents with experimentally chronicapical periodontitis, infection with ATCC29212 10 CFU was performed. In group III, the treatment group, after being injected with , 10 μL propolis was applied. It required 21 days to induce post-pulp chronic apical periodontitis infection. The rats were euthanised for immunohistochemical examination in order to measure the expression of OPG and to count histologically the number of osteoclast.
The expression of OPG and osteoclast constituted 17.5±1.58 and 6.4±0.96 in group I, 10±2 and 16.2±1.31 in group II and 17±1.69 and 7.5±1.08 in group III. Group I presented the highest level of OPG expression but the lowest level of osteoclast expression. There were significant differences between groups II and III and group I regarding OPG and osteoclast expression (<0.05).
East Java Propolis was a potential intracanal medicament promoting an increase in osteoprotegerin expression and a decrease in the number of osteoclasts thereby inhibiting osteoclastogenesis.
本动物研究的目的是通过评估骨保护素(OPG)的表达和破骨细胞水平,将东爪哇蜂胶作为治疗根尖慢性根尖周炎骨吸收的潜在天然根管内药物进行推广。
采用浸渍法制备蜂胶提取物。30只Wistar大鼠分为三组。第一组为对照组,右上第一磨牙为健康牙齿。第二组为实验性慢性根尖周炎组,用10CFU的ATCC29212进行感染。第三组为治疗组,注射后,应用10μL蜂胶。诱导牙髓后慢性根尖周炎感染需要21天。为了测量OPG的表达并组织学计数破骨细胞数量,对大鼠实施安乐死以进行免疫组织化学检查。
第一组中OPG的表达和破骨细胞数量分别为17.5±1.58和6.4±0.96,第二组为10±2和16.2±1.31,第三组为17±1.69和7.5±1.08。第一组OPG表达水平最高,但破骨细胞表达水平最低。第二组和第三组与第一组在OPG和破骨细胞表达方面存在显著差异(<0.05)。
东爪哇蜂胶是一种潜在的根管内药物,可促进骨保护素表达增加,减少破骨细胞数量,从而抑制破骨细胞生成。
