The currently available agents for use against toxoplasmosis have serious limitations. Thus, the aim of the present study was to investigate the anti- () activities of methanol extracts of () (leaves and fruits), () (fruits), and () (leaves) and .
Vero cells were treated with different concentrations (from 0 to 400 μg/mL) of the above extracts or with pyrimethamine at a dose of 50 mg/mL (positive control). Then, the viabilities of the -infected cells were measured by using colorimetric MTT (3-(4, 5-dimethylthiazol-2-yl) 2, 5-diphenyltetrazolium bromide) assays. In addition, the survival rates of mice acutely infected with 2 × 10 RH strain tachyzoites of were examined after intraperitoneal injection of the extracts at doses of 100 and 200 mg/kg/day for 5 days.
In the anti- assay, the IC values were 12.77, 180.2, 74.73, 213.2 and 163.8 μg/mL, and the selectivity indices were 6.05, 1.31, 0.35, 0.69 and 1.30 for the (leaves and fruits), , and extracts and pyrimethamine, respectively. Moreover, the mice treated with (leaves and fruits) achieved better results in terms of survival than the others ( < 0.05).
The results of the current study indicate that methanol extract of has significant anti- activity. Further study should be conducted to investigate the potential bioactivity of this extract through bioactivity-guided fractionation.
目前可用的抗弓形虫病药物存在严重局限性。因此,本研究的目的是调查[植物名称1](叶和果实)、[植物名称2](果实)和[植物名称3](叶)的甲醇提取物以及[提取物名称]的抗(弓形虫)活性。
用上述不同浓度(0至400μg/mL)的提取物或50mg/mL剂量的乙胺嘧啶(阳性对照)处理Vero细胞。然后,使用比色MTT(3-(4,5-二甲基噻唑-2-基)-2,5-二苯基四氮唑溴盐)测定法测量被弓形虫感染细胞的活力。此外,在以100和200mg/kg/天的剂量腹腔注射提取物5天后,检查急性感染2×10[弓形虫株名称]速殖子的小鼠的存活率。
在抗弓形虫试验中,[植物名称1](叶和果实)、[植物名称2]、[植物名称3]提取物以及乙胺嘧啶的IC值分别为12.77、180.2、74.73、213.2和163.8μg/mL,选择性指数分别为6.05、1.31、0.35、0.69和1.30。此外,用[植物名称1](叶和果实)处理的小鼠在存活率方面比其他组取得了更好的结果(P<0.05)。
本研究结果表明[植物名称1]的甲醇提取物具有显著的抗弓形虫活性。应通过生物活性导向分级分离进一步研究该提取物的潜在生物活性。
