A short rod-shaped, yellow-orange pigmented, strictly aerobic bacterium, designated as strain H2, was isolated from the wetland soil of Halla Mountain, Jeju-island, South Korea. Growth was observed at temperatures of 10-30 °C (optimum at 25-30 °C), pH of 6-8 (optimum at pH 7), and salt concentrations of 0-1% (w/v) NaCl (optimum at 0%). The strain H2 was found to be a catalase and oxidase-positive, non-motile, Gram-negative bacterium. On the basis of 16S rRNA gene sequence similarity and phylogenetic analysis, strain H2 was found to be related to the members of the Chitinophagaceae family, being closely related to Taibaiella chishuiensis AY17 (94.3% sequence similarity). The major polar lipids are phosphatidylethanolamine and glycolipid. Strain H2 contained MK-7 as the only menaquinone as well as iso-C, iso-C G and iso-C 3-OH as the major fatty acids (> 15%). The DNA G+C content of strain H2 was determined to be 48.3 mol%. Based on the phylogenetic, phenotypic characteristics and chemotaxonomic analysis data, strain H2 (= KCTC 62115 = JCM 32353) should be classified as representative of a novel species of a novel genus within the family Chitinophagaceae, for which the name Edaphocola aurantiacus gen. nov., sp. nov., is proposed.
从韩国济州岛汉拿山湿地土壤中分离出一株短杆状、黄色至橙色色素沉着、严格需氧的细菌,命名为H2菌株。该菌株在10-30°C(最适温度为25-30°C)、pH值为6-8(最适pH值为7)、盐浓度为0-1%(w/v)NaCl(最适浓度为0%)的条件下生长。H2菌株为过氧化氢酶和氧化酶阳性、无运动性、革兰氏阴性细菌。基于16S rRNA基因序列相似性和系统发育分析,发现H2菌株与几丁质噬菌科成员相关,与赤水太白杆菌AY17密切相关(序列相似性为94.3%)。主要极性脂类为磷脂酰乙醇胺和糖脂。H2菌株含有MK-7作为唯一的甲基萘醌,以及异-C、异-C G和异-C 3-OH作为主要脂肪酸(>15%)。H2菌株的DNA G+C含量测定为48.3 mol%。基于系统发育、表型特征和化学分类分析数据,H2菌株(=KCTC 62115=JCM 32353)应被归类为几丁质噬菌科内一个新属的新物种的代表,为此提出新属名Edaphocola aurantiacus gen. nov., sp. nov.。
