Abstract

A Gram-stain-negative, aerobic, catalase-positive, oxidase-positive, motile by a single polar flagellum, rod-shaped strain, designated DSW25-10, was isolated from the deep-sea water of the East Sea, Republic of Korea. Strain DSW25-10grew at 4-35 °C (optimum, 20-35 °C), at pH 5.5-9.0 (optimum, pH 7.0) and in the presence of 0-6.0 % NaCl (optimum, 0.5-2 %), and could assimilate valerate, but not assimilate d-mannitol and 3-hydroxy-butyrate. Comparative 16S rRNA gene sequence analysis showed that strain DSW25-10 belongs to the genus Zhongshania in the family Spongiibacteraceae and is most closely related to Zhongshania guokunii ZS6-22, Zhongshania borealis CL-AS9, Zhongshania aliphaticivorans SM-2 and Zhongshani antarctica ZS5-23 with a similarities of 97.1, 97.0, 96.7 and 96.6 %, respectively. The G+C content of genomic DNA was 49.1 mol% and the major respiratory quinone was ubiquinone-8. Phosphatidylethanolamine, phosphatidylglycerol and diphosphatidylglycerol were identified as the major cellular polar lipids. The predominant cellular fatty acids in strain DSW25-10 were summed feature 3 (comprising C16 : 1ω7c and/or C16 : 1ω6c), summed feature 8 (comprising C18 : 1ω7c and/or C18 : 1ω6c), C16 : 0 and C17 : 1ω8c. The DNA-DNA relatedness values between strain DSW25-10 and related strains were clearly lower than 70 %. On the basis of evidence from a polyphasic analysis, strain DSW25-10 is proposed to represent a novel species, Zhongshania marina sp. nov. The type strain is DSW25-10 (=KCCM 43273=JCM 17372).

从大韩民国东海的深海水中分离出一株革兰氏阴性、需氧、过氧化氢酶阳性、氧化酶阳性、具单极鞭毛运动的杆状菌株,命名为DSW25-10。菌株DSW25-10在4-35 °C(最适温度为20-35 °C)、pH 5.5-9.0(最适pH为7.0)以及0-6.0 % NaCl(最适浓度为0.5-2 %)的条件下生长,能同化戊酸,但不能同化d-甘露醇和3-羟基丁酸。16S rRNA基因序列比较分析表明,菌株DSW25-10属于海绵杆菌科中的中三亚属,与郭氏中三亚菌ZS6-22、北方中三亚菌CL-AS9、脂肪分解中三亚菌SM-2和南极中三亚菌ZS5-23关系最为密切,相似性分别为97.1%、97.0%、96.7%和96.6%。基因组DNA的G+C含量为49.1 mol%,主要呼吸醌为泛醌-8。磷脂酰乙醇胺、磷脂酰甘油和二磷脂酰甘油被鉴定为主要的细胞极性脂质。菌株DSW25-10中主要的细胞脂肪酸为总和特征3(包含C16 : 1ω7c和/或C16 : 1ω6c)、总和特征8(包含C18 : 1ω7c和/或C18 : 1ω6c)、C16 : 0和C17 : 1ω8c。菌株DSW25-10与相关菌株之间的DNA-DNA相关性值明显低于70 %。基于多相分析的证据,菌株DSW25-10被提议代表一个新物种,即海洋中三亚菌(Zhongshania marina sp. nov.)。模式菌株为DSW25-10(=KCCM 43273=JCM 17372)。
