Hypoglycemic Effects of Oat Oligopeptides in High-Calorie Diet/STZ-Induced Diabetic Rats.

Abstract

The study was aimed to determine whether treatment with oat oligopeptides (OOPs) could modulate hyperglycemia related to type 2 diabetes mellitus (T2DM) in Sprague⁻Dawley (SD) rats. Diabetic SD rats modeling by a joint effect of high-calorie diet for 45 days and twice intraperitoneal injection of 30 mg/kg streptozotocin at one-week interval were observed with or without OOPs administration (0.25, 0.50, 1.00, and 2.00 g/kg Body Weight) for 12 weeks. Fasting blood glucose (FBG), oral glucose test tolerance (OGTT), serum insulin, level of antioxidant, and hepatic enzymes were measured. In addition, frequency of micturition was recorded in this study for the first time. It was observed that the administration of OOPs (2.00 g/kg Body Weight) resulted in a significant decrease ( < 0.05) in FBG since 6th week and a significant decrease ( < 0.05) in the OGTT-AUC on 6th and 10th week. In addition, the administration of OOPs (2.00 g/kg Body Weight) reduced HOMA-IR index and 24-h urine volume significantly ( < 0.05) whereas increased SOD activity significantly ( < 0.05). These results suggested that OOPs may have a hypoglycemic effect in diabetic rats.

本研究旨在探讨燕麦低聚肽(OOPs)是否能调节 2 型糖尿病(T2DM)大鼠的高血糖。通过给予高糖饮食 45 天和间隔一周两次腹腔注射 30mg/kg 链脲佐菌素,建立 Sprague-Dawley(SD)大鼠糖尿病模型,观察或不给予 OOPs(0.25、0.5、1.0 和 2.0g/kg 体重)治疗 12 周。检测空腹血糖(FBG)、口服葡萄糖耐量试验(OGTT)、血清胰岛素、抗氧化和肝酶水平,同时记录首次排尿频率。结果发现,OOPs(2.0g/kg 体重)治疗 6 周后 FBG 显著降低(<0.05),第 6 周和第 10 周 OGTT-AUC 显著降低(<0.05)。此外,OOPs(2.0g/kg 体重)治疗还可显著降低 HOMA-IR 指数和 24 小时尿量(<0.05),同时显著提高 SOD 活性(<0.05)。这些结果表明,OOPs 可能对糖尿病大鼠具有降血糖作用。
