Strain ICH-30 was isolated from a freshwater fish pond in Taiwan. Phylogenetic analyses based on 16S rRNA gene sequences and coding sequences of 92 protein clusters indicated that strain ICH-30 formed a phylogenetic lineage in the genus Flavobacterium. Strain ICH-30 was most closely related to Flavobacteriumlacus NP180 with 97.0 % 16S rRNA gene sequence similarity. Strain ICH-30 showed 70.0-83.1 % average nucleotide identity and 16.5-23.7 % digital DNA-DNA hybridization identity with the type strains of other closely related Flavobacterium species. Cells of strain ICH-30 were Gram-stain-negative, strictly aerobic, non-motile, rod-shaped and formed dark orange colonies. Optimal growth occurred at 20-30 °C, pH 8-9 and 0-0.5 % NaCl. Strain ICH-30 contained iso-C15 : 1 G and iso-C15 : 0 as the predominant fatty acids. The major hydroxyl fatty acids were iso-C17 : 0 3-OH, iso-C16 : 0 3-OH and iso-C15 : 0 3-OH. The polar lipid profile consisted of phosphatidylethanolamine, six uncharacterized aminophospholipids, one uncharacterized phospholipid and two uncharacterized lipids. The major polyamine was homospermidine. The only isoprenoid quinone was MK-6. The DNA G+C content of the genomic DNA was 34.3 mol%. On the basis of the phylogenetic inference and phenotypic data, strain ICH-30 should be classified as a novel species, for which the name Flavobacterium piscinae sp. nov. is proposed. The type strain is ICH-30 (=BCRC 81122=LMG 30579=KCTC 62513).
菌株ICH-30是从台湾的一个淡水鱼塘中分离出来的。基于16S rRNA基因序列和92个蛋白质簇的编码序列进行的系统发育分析表明,菌株ICH-30在黄杆菌属中形成了一个系统发育谱系。菌株ICH-30与湖泊黄杆菌NP180的关系最为密切,16S rRNA基因序列相似性为97.0%。菌株ICH-30与其他密切相关的黄杆菌属模式菌株的平均核苷酸同一性为70.0-83.1%,数字DNA-DNA杂交同一性为16.5-23.7%。菌株ICH-30的细胞革兰氏染色阴性,严格需氧,不运动,杆状,形成深橙色菌落。最适生长温度为20-30°C,pH值为8-9,NaCl浓度为0-0.5%。菌株ICH-30以异-C15:1 G和异-C15:0作为主要脂肪酸。主要的羟基脂肪酸为异-C17:0 3-OH、异-C16:0 3-OH和异-C15:0 3-OH。极性脂质谱由磷脂酰乙醇胺、六种未鉴定的氨基磷脂、一种未鉴定的磷脂和两种未鉴定的脂质组成。主要的多胺是高亚精胺。唯一的类异戊二烯醌是MK-6。基因组DNA的G+C含量为34.3 mol%。根据系统发育推断和表型数据,菌株ICH-30应被分类为一个新物种,为此提出了新名称鱼黄杆菌(Flavobacterium piscinae sp. nov.)。模式菌株为ICH-30(=BCRC 81122=LMG 30579=KCTC 62513)。
