A novel Gram-stain-negative, strictly aerobic bacterium that has a rod-like shape with a single polar flagellum in the exponential phase of growth and a spherical or ovoid shape without a flagellum in the stationary phase was isolated from a mangrove wetland sediment sample collected at Beilun Estuary National Nature Reserve, Guangxi Province, PR China and designated strain ZS-4. This strain grew optimally at pH 6.0-8.0, at a temperature of 37 °C and in the presence of 3-4 % (w/v) NaCl. Its polar lipid profile included phosphatidylethanolamine, phosphatidylglycerol, one unidentified aminophospholipid and two uncharacterized lipids. Ubiquinone 8 (Q-8) was the sole respiratory quinone and the cellular fatty acids were dominated by Cω8 and C. A phylogenetic analysis based on the 16S rRNA gene sequence showed that strain ZS-4 exhibited its highest similarities to the type strains HMF4135 (97.8 %) and GJSW-36 (95.9 %). A whole genome-level comparison of strain ZS-4 with MCCC 1K03283 revealed an average nucleotide identity value of 75.6 % and a calculated DNA-DNA hybridization value of 19.6 %. In addition, the genomic DNA G+C content of strain ZS-4 was 45.9 mol%. Thus, based on analyses of its morphology, physiology, fatty acid composition and 16S rRNA gene sequence, strain ZS-4 should be considered a novel species of the genus , with the proposed name sp. nov. The type strain is ZS-4 (=KCTC 72399=MCCC 1K03630).
一株新的革兰氏阴性、严格需氧细菌,在指数生长期呈杆状,有一个单极鞭毛,在静止期呈球形或卵圆形,没有鞭毛。该菌从中国广西北仑河口国家自然保护区红树林湿地沉积物样品中分离得到,命名为菌株 ZS-4。该菌在 pH6.0-8.0、温度 37°C 和 3-4%(w/v)NaCl 条件下生长最佳。其极性脂谱包括磷脂酰乙醇胺、磷脂酰甘油、一种未鉴定的氨基磷脂和两种未鉴定的脂类。泛醌 8(Q-8)是唯一的呼吸醌,细胞脂肪酸主要由 Cω8 和 C 组成。基于 16S rRNA 基因序列的系统发育分析表明,菌株 ZS-4与模式菌株 HMF4135(97.8%)和 GJSW-36(95.9%)的相似度最高。与 MCCC 1K03283 的全基因组水平比较显示,菌株 ZS-4与 MCCC 1K03283 的平均核苷酸同一性值为 75.6%,计算的 DNA-DNA 杂交值为 19.6%。此外,菌株 ZS-4 的基因组 DNA G+C 含量为 45.9mol%。因此,基于形态学、生理学、脂肪酸组成和 16S rRNA 基因序列的分析,菌株 ZS-4 应被视为属的一个新种,命名为 sp. nov. 模式菌株为 ZS-4(=KCTC 72399=MCCC 1K03630)。
