A head injury is a very dangerous condition that threatens human life. This study examines the use of caffeic acid phenethyl ester (CAPE) in reducing cerebral edema in cases of head injury. The purpose of this study is to demonstrate whether CAPE can improve various parameters related to the expression of Aquaporin-4 (AQP4) mRNA and the serum AQP4 levels in rat subjects.
This is a randomized controlled study using a posttest-only control group design that uses experimental animals-specifically, male ( strain) rats aged 10-12 weeks and weighing 200-300 g. This study used a head injury model according to Marmarou (1994) with minor modifications to the animal model fixation tool. The parameters of the AQP4 mRNA were examined with real-time PCR, while serum AQP4 levels were examined with sandwich ELISA.
The AQP4 mRNA expression in rats that were given CAPE was lower than those not given CAPE, both on the fourth and seventh days; serum AQP4 levels in rats that were given CAPE were also lower than those not given CAPE, both on the fourth and seventh days.
Administration of CAPE in a rat model with head injury can reduce cerebral edema, mediated by AQP4.
头部损伤是一种威胁人类生命的非常危险的状况。本研究考察了咖啡酸苯乙酯(CAPE)在减轻头部损伤病例脑水肿方面的应用。本研究的目的是证明CAPE是否能改善与大鼠水通道蛋白-4(AQP4)mRNA表达及血清AQP4水平相关的各项参数。
这是一项随机对照研究,采用仅后测对照组设计,使用实验动物——具体为10 - 12周龄、体重200 - 300克的雄性(品系)大鼠。本研究采用根据Marmarou(1994)改良的头部损伤模型,对动物模型固定工具进行了微小改动。采用实时聚合酶链反应检测AQP4 mRNA参数,采用夹心酶联免疫吸附测定法检测血清AQP4水平。
给予CAPE的大鼠在第4天和第7天的AQP4 mRNA表达均低于未给予CAPE的大鼠;给予CAPE的大鼠在第4天和第7天的血清AQP4水平也低于未给予CAPE的大鼠。
在大鼠头部损伤模型中给予CAPE可减轻由AQP4介导的脑水肿。
